%22%3A%22S%5Cu00fcdhof%22%7D%5D%2C%22abstractNote%22%3A%22Heterozygous%20NRXN1%20deletions%20constitute%20the%20most%20prevalent%20currently%20known%20single-gene%20mutation%20associated%20with%20schizophrenia%2C%20and%20additionally%20predispose%20to%20multiple%20other%20neurodevelopmental%20disorders.%20Engineered%20heterozygous%20NRXN1%20deletions%20impaired%20neurotransmitter%20release%20in%20human%20neurons%2C%20suggesting%20a%20synaptic%20pathophysiological%20mechanism.%20Utilizing%20this%20observation%20for%20drug%20discovery%2C%20however%2C%20requires%20confidence%20in%20its%20robustness%20and%20validity.%20Here%2C%20we%20describe%20a%20multicenter%20effort%20to%20test%20the%20generality%20of%20this%20pivotal%20observation%2C%20using%20independent%20analyses%20at%20two%20laboratories%20of%20patient-derived%20and%20newly%20engineered%20human%20neurons%20with%20heterozygous%20NRXN1%20deletions.%20Using%20neurons%20transdifferentiated%20from%20induced%20pluripotent%20stem%20cells%20that%20were%20derived%20from%20schizophrenia%20patients%20carrying%20heterozygous%20NRXN1%20deletions%2C%20we%20observed%20the%20same%20synaptic%20impairment%20as%20in%20engineered%20NRXN1-deficient%20neurons.%20This%20impairment%20manifested%20as%20a%20large%20decrease%20in%20spontaneous%20synaptic%20events%2C%20in%20evoked%20synaptic%20responses%2C%20and%20in%20synaptic%20paired-pulse%20depression.%20Nrxn1-deficient%20mouse%20neurons%20generated%20from%20embryonic%20stem%20cells%20by%20the%20same%20method%20as%20human%20neurons%20did%20not%20exhibit%20impaired%20neurotransmitter%20release%2C%20suggesting%20a%20human-specific%20phenotype.%20Human%20NRXN1%20deletions%20produced%20a%20reproducible%20increase%20in%20the%20levels%20of%20CASK%2C%20an%20intracellular%20NRXN1-binding%20protein%2C%20and%20were%20associated%20with%20characteristic%20gene-expression%20changes.%20Thus%2C%20heterozygous%20NRXN1%20deletions%20robustly%20impair%20synaptic%20function%20in%20human%20neurons%20regardless%20of%20genetic%20background%2C%20enabling%20future%20drug%20discovery%20efforts.%22%2C%22date%22%3A%222021%5C%2F06%5C%2F01%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1073%5C%2Fpnas.2025598118%22%2C%22ISSN%22%3A%220027-8424%2C%201091-6490%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fwww.pnas.org%5C%2Fcontent%5C%2F118%5C%2F22%5C%2Fe2025598118%22%2C%22deleted%22%3A1%2C%22collections%22%3A%5B%22TUURQW3H%22%5D%2C%22dateModified%22%3A%222021-05-31T18%3A08%3A56Z%22%7D%7D%5D%7D
