%20human-derived%20organoids%20treated%20with%20the%20cannabinoid%20agonist%20WIN%2055%2C212-2%20exhibited%20the%20least%20convergence%20of%20all%20groups.%20Single-cell%20analysis%20revealed%20that%20WIN%2055%2C212-2%20increased%20DNA%20fragmentation%2C%20an%20indicator%20of%20apoptosis%2C%20in%20human-derived%20dorsal%20forebrain%20organoids.%20We%20subsequently%20confirmed%20induction%20of%20DNA%20damage%20and%20apoptosis%20by%20WIN%2055%2C212-2%20within%203D%20human-derived%20dorsal%20forebrain%20organoids.%20Lastly%2C%20in%20a%20BrdU%20pulse-chase%20neocortical%20neurogenesis%20paradigm%2C%20we%20identified%20that%20WIN%2055%2C212-2%20was%20the%20only%20enviromimetic%20treatment%20to%20disrupt%20newborn%20neuron%20numbers%20within%20human-derived%20dorsal%20forebrain%20organoids.%20Cumulatively%20this%20study%20serves%20as%20both%20a%20resource%20and%20foundation%20from%20which%20human%203D%20biologics%20can%20be%20used%20to%20resolve%20the%20non-genomic%20effects%20of%20neuropsychiatric%20risk%20factors%20under%20controlled%20laboratory%20conditions.%20While%20synthetic%20cannabinoids%20can%20differ%20from%20naturally%20occurring%20compounds%20in%20their%20effects%2C%20our%20data%20nonetheless%20suggests%20that%20exposure%20to%20WIN%2055%2C212-2%20elicits%20neurotoxicity%20within%20human-derived%20developing%20forebrain%20tissue.%20These%20human-derived%20data%20therefore%20support%20the%20long-standing%20belief%20that%20maternal%20use%20of%20cannabinoids%20may%20require%20caution%20so%20to%20avoid%20any%20potential%20neurodevelopmental%20effects%20upon%20developing%20offspring%20in%20utero.%22%2C%22date%22%3A%222021-06-22%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1038%5C%2Fs41380-021-01189-9%22%2C%22ISSN%22%3A%221476-5578%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fwww.nature.com%5C%2Farticles%5C%2Fs41380-021-01189-9%22%2C%22deleted%22%3A1%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-06-22T16%3A21%3A44Z%22%7D%7D%5D%7D
