0erb-b2%20receptor%20tyrosine%20kinase%202%20%28HER2%29-specific%20CAR%20T%20cells%20in%20an%20orthotopic%20xenograft%20medulloblastoma%20model.%20We%20translated%20these%20findings%20into%20BrainChild-01%20%28NCT03500991%29%2C%20an%20ongoing%20phase%201%20clinical%20trial%20at%20Seattle%20Children%5Cu2019s%20evaluating%20repetitive%20locoregional%20dosing%20of%20these%20HER2-specific%20CAR%20T%20cells%20to%20children%20and%20young%20adults%20with%20recurrent%5C%2Frefractory%20CNS%20tumors%2C%20including%20diffuse%20midline%20glioma.%20Primary%20objectives%20are%20assessing%20feasibility%2C%20safety%20and%20tolerability%3B%20secondary%20objectives%20include%20assessing%20CAR%20T%20cell%20distribution%20and%20disease%20response.%20In%20the%20outpatient%20setting%2C%20patients%20receive%20infusions%20via%20CNS%20catheter%20into%20either%20the%20tumor%20cavity%20or%20the%20ventricular%20system.%20The%20initial%20three%20patients%20experienced%20no%20dose-limiting%20toxicity%20and%20exhibited%20clinical%2C%20as%20well%20as%20correlative%20laboratory%2C%20evidence%20of%20local%20CNS%20immune%20activation%2C%20including%20high%20concentrations%20of%20CXCL10%20and%20CCL2%20in%20the%20cerebrospinal%20fluid.%20This%20interim%20report%20supports%20the%20feasibility%20of%20generating%20HER2-specific%20CAR%20T%20cells%20for%20repeated%20dosing%20regimens%20and%20suggests%20that%20their%20repeated%20intra-CNS%20delivery%20might%20be%20well%20tolerated%20and%20activate%20a%20localized%20immune%20response%20in%20pediatric%20and%20young%20adult%20patients.%22%2C%22date%22%3A%222021-07-12%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1038%5C%2Fs41591-021-01404-8%22%2C%22ISSN%22%3A%221546-170X%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fwww.nature.com%5C%2Farticles%5C%2Fs41591-021-01404-8%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-07-12T18%3A19%3A54Z%22%7D%7D%5D%7D
