Secretome Screening Reveals Immunomodulating Functions of IFNα-7, PAP and GDF-7 on Regulatory T-cells

are%20highly%20interesting%20for%20both%20therapeutic%20and%20diagnostic%20purposes%20in%20either%20hyperactive%20or%20immunosuppressed%20populations.%20Here%2C%20we%20report%20a%20phenotypic%20screening%20of%20a%20human%20secretome%20library%20in%20human%20Treg%20cells%20utilising%20a%20high%20throughput%20flow%20cytometry%20technology.%20Screening%20a%20library%20of%20575%20secreted%20proteins%20allowed%20us%20to%20identify%20proteins%20stabilising%20or%20destabilising%20the%20Treg%20phenotype%20as%20suggested%20by%20changes%20in%20expression%20of%20Treg%20marker%20proteins%20FOXP3%20and%5C%2For%20CTLA4.%20Four%20proteins%20including%20GDF-7%2C%20IL-10%2C%20PAP%20and%20IFN%5Cu03b1-7%20were%20identified%20as%20positive%20regulators%20that%20increased%20FOXP3%20and%5C%2For%20CTLA4%20expression.%20PAP%20is%20a%20phosphatase.%20A%20catalytic-dead%20version%20of%20the%20protein%20did%20not%20induce%20an%20increase%20in%20FOXP3%20expression.%20Ten%20interferon%20proteins%20were%20identified%20as%20negative%20regulators%20that%20reduced%20the%20expression%20of%20both%20CTLA4%20and%20FOXP3%2C%20without%20affecting%20cell%20viability.%20A%20transcriptomics%20analysis%20supported%20the%20differential%20effect%20on%20Tregs%20of%20IFN%5Cu03b1-7%20versus%20other%20IFN%5Cu03b1%20proteins%2C%20indicating%20differences%20in%20JAK%5C%2FSTAT%20signaling.%20A%20conformational%20model%20experiment%20confirmed%20a%20tenfold%20reduction%20in%20IFNAR-mediated%20ISG%20transcription%20for%20IFN%5Cu03b1-7%20compared%20to%20IFN%5Cu03b1-10.%20This%20further%20strengthened%20the%20theory%20of%20a%20shift%20in%20downstream%20messaging%20upon%20external%20stimulation.%20As%20a%20summary%2C%20we%20have%20identified%20four%20positive%20regulators%20of%20FOXP3%20and%5C%2For%20CTLA4%20expression.%20Further%20exploration%20of%20these%20Treg%20modulators%20and%20their%20method%20of%20action%20has%20the%20potential%20to%20aid%20the%20discovery%20of%20novel%20therapies%20for%20both%20autoimmune%20and%20infectious%20diseases%20as%20well%20as%20for%20cancer.%22%2C%22date%22%3A%222021-08-18%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1038%5C%2Fs41598-021-96184-z%22%2C%22ISSN%22%3A%222045-2322%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fwww.nature.com%5C%2Farticles%5C%2Fs41598-021-96184-z%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-08-24T22%3A09%3A54Z%22%7D%7D%5D%7D
