Using patient-derived primary skin skin squamous cell carcinoma (SCC) cells or immortalized lines, the authors demonstrated that RNA polymerase mitochondrial (POLRMT) shRNA or knockout (KO) potently suppressed mitochondrial DNA (mtDNA) transcription and suppressed cell viability, proliferation and migration.
