2creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Eline%20T.%20Luning%22%2C%22lastName%22%3A%22Prak%22%7D%2C%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Paul%22%2C%22lastName%22%3A%22Bates%22%7D%2C%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Scott%20E.%22%2C%22lastName%22%3A%22Hensley%22%7D%2C%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Allison%20R.%22%2C%22lastName%22%3A%22Greenplate%22%7D%2C%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22E.%20John%22%2C%22lastName%22%3A%22Wherry%22%7D%5D%2C%22abstractNote%22%3A%22Novel%20mRNA%20vaccines%20for%20SARS-CoV-2%20have%20been%20authorized%20for%20emergency%20use.%20Despite%20their%20efficacy%20in%20clinical%20trials%2C%20data%20on%20mRNA%20vaccine-induced%20immune%20responses%20are%20mostly%20limited%20to%20serological%20analyses.%20Here%2C%20we%20interrogated%20antibody%20and%20antigen-specific%20memory%20B%20cells%20over%20time%20in%2033%20SARS-CoV-2%20na%5Cu00efve%20and%2011%20SARS-CoV-2%20recovered%20subjects.%20SARS-CoV-2%20na%5Cu00efve%20individuals%20required%20both%20vaccine%20doses%20for%20optimal%20increases%20in%20antibodies%2C%20particularly%20for%20neutralizing%20titers%20against%20the%20B.1.351%20variant.%20Memory%20B%20cells%20specific%20for%20full-length%20spike%20protein%20and%20the%20spike%20receptor%20binding%20domain%20%28RBD%29%20were%20also%20efficiently%20primed%20by%20mRNA%20vaccination%20and%20detectable%20in%20all%20SARS-CoV-2%20naive%20subjects%20after%20the%20second%20vaccine%20dose%2C%20though%20the%20memory%20B%20cell%20response%20declined%20slightly%20with%20age.%20In%20SARS-CoV-2%20recovered%20individuals%2C%20antibody%20and%20memory%20B%20cell%20responses%20were%20significantly%20boosted%20after%20the%20first%20vaccine%20dose%3B%20however%2C%20there%20was%20no%20increase%20in%20circulating%20antibodies%2C%20neutralizing%20titers%2C%20or%20antigen-specific%20memory%20B%20cells%20after%20the%20second%20dose.%20This%20robust%20boosting%20after%20the%20first%20vaccine%20dose%20strongly%20correlated%20with%20levels%20of%20pre-existing%20memory%20B%20cells%20in%20recovered%20individuals%2C%20identifying%20a%20key%20role%20for%20memory%20B%20cells%20in%20mounting%20recall%20responses%20to%20SARS-CoV-2%20antigens.%20Together%2C%20our%20data%20demonstrated%20robust%20serological%20and%20cellular%20priming%20by%20mRNA%20vaccines%20and%20revealed%20distinct%20responses%20based%20on%20prior%20SARS-CoV-2%20exposure%2C%20whereby%20COVID-19%20recovered%20subjects%20may%20only%20require%20a%20single%20vaccine%20dose%20to%20achieve%20peak%20antibody%20and%20memory%20B%20cell%20responses.%20These%20findings%20also%20highlight%20the%20utility%20of%20defining%20cellular%20responses%20in%20addition%20to%20serologies%20and%20may%20inform%20SARS-CoV-2%20vaccine%20distribution%20in%20a%20resource-limited%20setting.%5CnKey%20differences%20in%20both%20antibody%20and%20memory%20B%20cell%20responses%20to%20mRNA%20vaccination%20were%20identified%20based%20on%20prior%20SARS-CoV-2%20exposure.%5CnKey%20differences%20in%20both%20antibody%20and%20memory%20B%20cell%20responses%20to%20mRNA%20vaccination%20were%20identified%20based%20on%20prior%20SARS-CoV-2%20exposure.%22%2C%22date%22%3A%222021%5C%2F04%5C%2F15%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1126%5C%2Fsciimmunol.abi6950%22%2C%22ISSN%22%3A%222470-9468%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fimmunology.sciencemag.org%5C%2Fcontent%5C%2F6%5C%2F58%5C%2Feabi6950%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-06-14T18%3A09%3A10Z%22%7D%7D%5D%7D
