Soluble TREM2 Engages Cell-Surface Nucleolin to Drive Vascular Permeability and Malignant Ascites in Ovarian Cancer

[EMBO Molecular Medicine] Using a combination of in vitro, in vivo, and ex vivo models, scientists demonstrated that soluble TREM2 (sTREM2) promotes vascular permeability and drives ascites formation. Mechanistically, sTREM2 binds to nucleolin on endothelial cells and activates the AKT/eNOS signaling pathway, which leads to VE-cadherin phosphorylation and ultimately vascular leakage.

Vascular Mechanical Forces and Vascular Diseases

[Journal of Advanced Research] While the significance of mechanical forces in vascular diseases is widely acknowledged, substantial gaps persist in understanding how these forces are sensed and transduced in endothelial cells and vascular smooth muscle cells, as well as their involvement in various vascular diseases.

Four MCG Early-Career Researchers Secure AHA Funding

[Jagwire] Four early-career researchers from the Medical College of Georgia at Augusta University have been recently awarded Bridge Career Development Awards from the American Heart Association.

EZH2 Inhibition via GSK-126 Mitigates EndMT and Atherosclerosis in Diabetes: A Translational Epigenetic Approach

[Science Advances] Researchers showed that enhancer of zeste homolog 2 (EZH2)-mediated histone H3 at lysine-27 trimethylation is elevated in carotid plaques from patients with diabetes and in the aortic endothelium of diabetic Apoe−/− mice. They identified EZH2 as a driver of endothelial-to-mesenchymal transition in diabetes-associated atherosclerosis.

Cigarette Smoke-Induced Endothelial Senescence-Like Phenotype Promotes Lung Adenocarcinoma Metastasis via the BMP4-Smad1/5/9-ID1 Axis

[Ecotoxicology and Environmental Safety] Investigators hypothesized that cigarette smoke induces endothelial cell senescence, leading to the secretion of bone morphogenetic protein 4 (BMP4), which subsequently disrupts the endothelial barrier and promotes lung adenocarcinoma metastasis by activating the p-Smad1/5/9-ID1 signaling axis to drive epithelial-mesenchymal transition-like changes.

Mitochondrial Activity Promotes Neutrophil Degranulation and Endothelial Dysfunction in Systemic Infections

[EMBO Molecular Medicine] The authors showed that neutrophil degranulation can induce endothelial dysfunction via degradation of the glycocalyx and increase of endothelial permeability. They provide a mechanistic explanation for the hyperinflammatory nature of immature neutrophils.

Enhancers That Direct Gene Expression to Central Nervous System Vascular Endothelial Cells In Vivo

[Neuron] Researchers interrogated the function of putative cis-regulatory elements in brain endothelial cells (ECs) using transgenic mice and in vivo viral transduction of CNS ECs. The results provide evidence for functional enhancer and repressor elements and demonstrate the brain EC-specificity of one enhancer located upstream of the gene coding for GLUT1.

Identification of Unannotated Microproteins Involved in Endothelial Cell Homeostasis, Dysfunction, and Vascular Disease

[Cardiovascular Research] Investigators studied heart, lung, and blood vessels from endothelial cell-specific RiboTag mice and human endothelial cells under homeostatic and inflammatory conditions. They demonstrate the relevance of 250 microproteins for endothelial cell growth and viability.

Flow-Mediated Endothelial Remodeling and Inflammation Drive Developmental Vascular Susceptibility in Ldlr Loss of Function

[Nature Communications] Single-cell RNA-sequencing under differential flow conditions in embryonic low-density lipoprotein receptor knockout (ldlr-/-) zebrafish identified a population of disproportionately stressed endothelial cells marked by overexpression of heat shock protein 70.

Procr+ Endothelial Progenitor Cells Govern Hematopoiesis through Fine-Tuning Mesenchymal Stem Cell Niche Signals

[Blood] Scientists identified a subset of Procr+ endothelial cells (ECs) with progenitor-like properties in bone marrow. Procr binds HSPA8 to promote Foxc2 nuclear translocation, upregulating Dll4 transcription to sustain Dll4/Notch3 activation in MSCs, revealing a Procr/HSPA8/Foxc2/Dll4 axis essential for EC and MSC crosstalk.

CreER Activation Transiently Disrupts Angiogenesis by Reducing Proliferation and Promoting Apoptosis in Vascular Endothelial Cells

[Angiogenesis] Scientists demonstrated that ubiquitous CreER activation in postnatal mice decreases body growth and impairs retinal angiogenesis. They further showed that CreER activation slows endothelial cell proliferation and promotes apoptosis concomitant with p21/CDKN1A upregulation in the angiogenic retina.

Engineering and Integrating Blood Vessel Organoids: A Comprehensive Blueprint for Physiological and Disease Modeling

[Stem Cell Reviews and Reports] The authors sumarize methods for in vitro blood vessel generation and explore how vascularizing parenchymal organoids actively drives tissue maturation while overcoming hypoxic limitations.
