Hydrogel for Sustained and Localized Delivery of a Regulatory T Cell-Inducing Agent Promotes Bone Regeneration
[Nature Biomedical Engineering] Researchers engineered a hydrogel-based system for in situ Treg activation at injury sites through the controlled release of AS2863619, a cyclin-dependent kinase inhibitor.
Viral Subversion of B Cell Tolerance: Mechanisms Linking Infection to Autoimmunity
[EMBO Reports] The authors propose that autoimmune outcomes are determined less by viral identity than by the immunological and anatomical context in which tolerance fails.
Treg-Associated Oligomannose N-Glycans Trigger Immune Dysregulation in Myasthenia Gravis
[Journal of Autoimmunity] Scientists analyzed the surface N-glycosylation pattern in Tregs from Myasthenia gravis (MG) patients. They demonstrated in vitro that the glycoreprogramming of Tregs, mimicking the glycan composition found in MG patients, leads to decreased suppression function.
Immutep and Monash University Secure Further ARC Linkage Projects Funding for LAG-3 Research
[Immutep Limited] Immutep Limited announced that the Australian Research Council (ARC) has awarded Monash University a grant of A$652,248 over three years under the ARC’s Linkage Projects scheme to support its continuing research collaboration with Immutep into Lymphocyte Activation Gene-3 (LAG-3).
Glycome Remodeling Programs Pathogenic γδ T17 Cell Responses in Autoimmunity
[Science Immunology] Analysis of human transcriptomic data and murine models revealed that the absence of β1,6-N-acetylglucosamine–branched N-glycans promoted type 3 IL-17A–producing γδ T cells and loss of tolerance.
AIM2 in Tregs Restrains Anti-PD-1 Efficacy by Reinforcing Immune Suppression in MC38 Colorectal Cancer
[iScience] Investigators showed that the DNA sensor absent in melanoma 2 (AIM2) sustains immunosuppressive programs in Tregs and limits responses to immune checkpoint blockade. Using a colorectal cancer model, Treg specific deletion of AIM2 enhanced the efficacy of PD1 blockade.
Tregs Transduced with a Ro60-Reactive TCR Have Increased Immunosuppressive Functions in Sjögren’S Disease
[iScience] Scientists presented the first preclinical evidence supporting a TCR-engineered Treg (TCR-Treg) therapy specifically targeting Sjögren’s disease. They designed a TCR-Treg that recognizes a dominant epitope of the Ro60 antigen presented by HLA-DRB1∗03:01 and demonstrate that these cells can selectively suppress autoreactive T cells more effectively than polyclonal Treg.
Multiple Sclerosis-Associated CD226 Variant Disrupts Vitamin D-Induced Type I Treg Differentiation by Impairing LFA-1 Activation
[Journal of Clinical Investigation] Researchers indicated that CD226 is pivotal for the generation of type I regulatory cells in the context of CD46 costimulation and vitamin D exposure. They highlight how genetic and environmental factors may exert a combined control over the tuning of T cell function in the context of multiple sclerosis and potentially other pathologies with immune dysregulation.
Autoimmunity Induced by Epigenome Alteration Affecting Thymic Treg Cell Development
[Proceedings of the National Academy of Sciences of the United States of America] Investigators showed that methyl-CpG-binding domain protein 3 deficiency in the T cell compartment resulted in spontaneous development of T cell–mediated multiorgan autoimmune diseases including fatal autoimmune myocarditis.
Zag Bio Advances ZAG-101 into Clinical Development for Type 1 Diabetes
[Zag Bio™, Inc.] Zag Bio™, Inc. announced it has advanced its lead program for type 1 diabetes, ZAG-101, into clinical development. The company’s first-in-human Phase Ib/IIa clinical trial will evaluate the safety, tolerability, pharmacokinetics and pharmacodynamics of ZAG-101 and explore its potential to preserve insulin-producing beta-cell function.
Lupus Research Alliance Awards $3 Million to Transform How Lupus Treatment Response is Predicted and Personalized
[LRA] The Lupus Research Alliance (LRA), announced the 2026 Global Team Science Award to Dr. Ioannis Parodis and his team of international multidisciplinary experts. The team will use the grant to develop predictive tools that help identify which treatments are most likely to work for people living with lupus, forecast disease progression, and detect early signs of flares.
Tim-3 Sustains Tumor Treg Stability and Function, Limiting Checkpoint Blockade Therapy Efficacy
[Cancer Immunology Research] Researchers showed that T cell immunoglobulin and mucin 3 (Tim-3) expression on tumor Tregs is required for the function and survival of these cells, in part through Akt and FOXO1 signaling.
