    A Red-Shifted Fluorescent Substrate for Aldehyde Dehydrogenase
    Researchers report a red-shifted fluorescent substrate for aldehyde dehydrogenase (ALDH), AldeRed 588-A, for labeling viable ALDHpos cells. They demonstrated that AldeRed 588-A successfully isolates ALDHhi human hematopoietic stem cells from heterogeneous cord blood mononuclear cells. [Nat Commun] Abstract

    Sphingosine Kinase 1 Expressed by Endothelial Colony-Forming Cells Has a Critical Role in Their Revascularization Activity
    Conditioned medium from human bone marrow-derived mesenchymal stem cells increased the angiogenic activity of cord blood endothelial colony-forming cells (ECFCs) in vitro (proliferation, migration, and pseudo-tube formation), the survival of ECFCs in mice (Matrigel Plug assay) and the capacity of ECFCs to promote the recovery of blood perfusion in mice with hindlimb ischemia.     Scientists investigated the role of netrin-1 in promoting the motility of human umbilical cord blood derived mesenchymal stem cells and its related signaling pathways. [Stem Cells Dev] Abstract

    Relationship of BK Polyoma Virus (BKV) in the Urine with Hemorrhagic Cystitis and Renal Function in Recipients of T-Cell Depleted Peripheral Blood and Cord Blood Stem Cell Transplants
    Researchers prospectively monitored 98 hematopoietic stem cell transplants by serial BKV PCR in the urine through Day (D) +100 to analyze the relationship between BKV viruria and hemorrhagic cystitis, serum creatinine and creatinine clearance through D +180 or death. Patients, median age 52 years, range 20-73, received T-cell depleted or cord blood allografts. [Biol Blood Marrow Transplant] Abstract

    Increased Migration of Human Mesenchymal Stromal Cells by Autocrine Motility Factor (AMF) Resulted in Enhanced Recruitment towards Hepatocellular Carcinoma
    Researchers analyzed AMF involvement on mesenchymal stromal cell (MSC) migration towards human hepatocellular carcinoma. The effects of AMF on MSCs from different sources (bone marrow, adipose tissue and perivascular cells from umbilical cord) were analyzed using in vitro migration assay; metalloproteinase 2 activity and expression of critical genes were studied by zymography and qRT-PCR, respectively.     Umbilical Cord Tissue-Derived Mesenchymal Stem Cells Induce Apoptosis in PC-3 Prostate Cancer Cells through Activation of JNK and Downregulation of PI3K/AKT Signaling
    Investigators elucidated the antitumor activity of human umbilical cord mesenchymal stem cells (hUCMSCs) in PC-3 prostate cancer cells in vitro and in vivo. Antitumor effects of UCMSCs on tumor growth were evaluated in a co-culture condition with PC-3 prostate cancer cells. PC-3 cells were subcutaneously (sc) injected into the left flank of nude mice and UCMSCs were sc injected into the right flank of the same mouse. [Stem Cell Res Ther] Abstract | Full Article

    Extracellular O-Linked N-Acetylglucosamine Is Enriched in Stem Cells Derived from Human Umbilical Cord Blood
    Researchers were able to show by immunological assays that human umbilical cord blood-derived mesenchymal stromal cells display O-linked N-acetylglucosamine (O-GlcNAc), the product of epidermal growth factor domain-specific O-linked GlcNAc transferase, and that O-GlcNAc is further elongated with galactose to form O-linked N-acetyllactosamine.
