    TOP STORY

Researchers Find Solution to Cell Death Problem Vexing Stem Cell Research
Researchers show that a molecular motor, called “nonmuscle myosin II,” which exists naturally inside each human pluripotent stem (hPS) cell and controls various cellular functions, triggers the death of hPS cells when they are broken down to single cells. [Press release from the University of California, Riverside discussing online prepublication in Nature Communications]

CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal)

Non-Muscle Myosin II Regulates Survival Threshold of Pluripotent Stem Cells
Researchers demonstrate that treatment with a specific inhibitor of non-muscle myosin II, blebbistatin, enhances the survival of human pluripotent stem cells under clonal density and suspension conditions, and, in combination with a synthetic matrix, supports a fully defined environment for self-renewal. [Nat Commun]

Generation of Rat Pancreas in Mouse by Interspecific Blastocyst Injection of Pluripotent Stem Cells
To examine the potential for xenogenic approaches in blastocyst complementation, researchers injected mouse or rat pluripotent stem cells (PSCs) into rat or mouse blastocysts, respectively, generating interspecific chimeras and thus confirming that PSCs can contribute to xenogenic development between mouse and rat. Stat3 Activation Is Limiting for Reprogramming to Ground State Pluripotency
Findings indicate that Jak/Stat3 participate directly in molecular reprogramming and that activation of this pathway is a limiting component. [Cell Stem Cell]

Myc Represses Primitive Endoderm Differentiation in Pluripotent Stem Cells
Researchers report that codeletion of c- and N-MYC in induced pluripotent stem cells and embryonic stem cells results in their spontaneous differentiation to primitive endoderm. [Cell Stem Cell]

Functional Diversity of ESC-Derived Motor Neuron Subtypes Revealed Through Intraspinal Transplantation
Current methods to examine and regulate the functional integration and plasticity of human embryonic stem cell (hESC)-derived neurons are cumbersome and technically challenging. Researchers engineered hESCs and their derivatives to express the light-gated Channelrhodopsin-2 protein to overcome these deficiencies. [Cell Stem Cell]

Functional Control of Transplantable Human ESC-Derived Neurons via Optogenetic Targeting
Researchers engineered human embryonic stem cells (hESCs) and their derivatives to express the light-gated Channelrhodopsin-2 protein to overcome deficiencies in current methods to examine and regulate the functional integration and plasticity of hESC-derived neurons. [Stem Cells]

High-Efficiency Induction of Neural Conversion in hESCs and hiPSCs with a Single Chemical Inhibitor of TGF-Beta Superfamily Receptors
Researchers identified compound C (also denoted as dorsomorphin), a protein kinase inhibitor, as a potent regulator of human embryonic stem cell (hESC) and human induced pluripotent stem cell (hiPSC) fate decisions. [Stem Cells]

The Wnt/Beta-Catenin Signaling Pathway Tips the Balance Between Apoptosis and Reprogramming of Cell-Fusion Hybrids
The present study provides evidence that spontaneous fusion occurs, while the survival of the reprogrammed clones is strictly dependent upon induction of a Wnt-mediated reprogramming pathway. [Stem Cells]

Molecular Signature of Primary Retinal Pigment Epithelium and Stem Cell-Derived RPE Cells
Results establish a panel of molecular markers for evaluating the fidelity of human pluripotent stem cell to retinal pigment epithelium (RPE) conversion. [Hum Mol Genet]

Long-Term Human Pluripotent Stem Cell Self-Renewal on Synthetic Polymer Surfaces
Researchers identified several polymers that can support self-renewal of human pluripotent stem cells (hPSCs). While most of these polymers provide support for only a short period of time, they identified a synthetic polymer poly(methyl vinyl ether-alt-maleic anhydride) that supported the long-term attachment, proliferation and self-renewal of HUES1, HUES9, and iPSCs.
