    Activity of the Type II JAK2 Inhibitor CHZ868 in B Cell Acute Lymphoblastic Leukemia
    To improve the efficacy of JAK2 inhibition in B cell acute lymphoblastic leukemia (B-ALL), scientists developed the type II inhibitor CHZ868, which stabilizes JAK2 in an inactive conformation. CHZ868 potently suppressed the growth of CRLF2-rearranged human B-ALL cells, abrogated JAK2 signaling, and improved survival in mice with human or murine B-ALL. [Cancer Cell] Abstract | Press Release

    Production of IL-10 by CD4+ Regulatory T Cells during the Resolution of Infection Promotes the Maturation of Memory CD8+ T Cells
    Researchers found that interleukin 10 (IL-10) derived from CD4+ regulatory T cells was necessary for the maturation of memory CD8+ T cells following acute infection with lymphocytic choriomeningitis virus. [Nat Immunol] Abstract

    Steatohepatitis as Impaired Immune Metabolism Induced by Liver Resident Regulatory T-Cell Depletion
    Investigators found that steatohepatitis was induced under the condition of temporary depletion of liver-resident regulatory T (Treg) cells combined with a high-fat diet. With the temporary depletion of Treg cells, interferon γ-secreting CD4+ T cells and CD8+ T cells were increased. [Hepatology] Abstract

    Depletion of B Cells Induce Remission of Autoimmune Hepatitis in Mice through Reduced Antigen Presentation and Help to T Cells
    Scientists aimed to understand the immunological changes underlying the autoimmune hepatitis (AIH) remission caused by B-cell depletion in an experimental model of AIH. [Hepatology] Abstract

    Opposing Roles of STAT1 and STAT3 in IL-21 Function in CD4+ T Cells
    Researchers investigated the role of STAT1 and showed that STAT1 and STAT3 have at least partially opposing roles in IL-21 signaling in CD4+ T cells. IL-21 induced STAT1 phosphorylation, and this was augmented in Stat3-deficient CD4+ T cells. [Proc Natl Acad Sci USA] Abstract

    Self-Recognition Drives the Preferential Accumulation of Promiscuous CD4 T-Cells in Aged Mice
    Scientists report that, while old mice contain fewer CD4+ T-cells compared with adults, those that remain have a higher intrinsic affinity for self-major histocompatibility complex molecules, as measured by CD5 expression. [Elife] Full Article

    Selective Involvement of the Checkpoint Regulator VISTA in Suppression of B-Cell, but Not T-Cell, Responsiveness by Monocytic Myeloid Derived Suppressor Cells from Mice Infected by an Immunodeficiency-Causing Retrovirus
    Researchers demonstrated augmentation of monocytic myeloid-derived suppressor cells whose suppression of not only T-, but also B-cell, responsiveness paralleled the immunodeficiency during LP-BM5 retrovirus infection. [J Virol] Abstract

    Transcription Factor IRF8 Controls Th1-Like Regulatory T-Cell Function
    Scientists discovered that transcription factor interferon regulatory factor 8 (IRF8) was selectively expressed in regulatory T and Th1 cells. IRF8-deficient Treg cells showed defective expression of CXCR3 and aberrant expression of the Il4 and Il17 genes. [Cell Mol Immunol] Abstract

    PARP Inhibition by Olaparib or Gene Knockout Blocks Asthma-Like Manifestation in Mice by Modulating CD4+ T Cell Function
    Investigators examined the efficacy of olaparib in blocking established allergic airway inflammation and hyperresponsiveness similar to those observed in human asthma in animal models of the disease. [J Transl Med] Full Article

    Pre-Vaccination Frequencies of Th17 Cells Correlate with Vaccine-Induced T-Cell Responses to Survivin-Derived Peptide Epitopes
    Scientists performed an exploratory analysis of a possible correlation of pre-vaccination Th17 cells, myeloid derived suppressor cells , and regulatory T cells with both vaccination-induced T-cell responses as well as clinical outcome in metastatic melanoma patients vaccinated with survivin-derived peptides.
