SCIENCE NEWS New Clues to How Cancer-Related Proteins Plasmin, Thrombin Lose Inhibition A new technique that searches blood for the tiniest remnants of broken down proteins has revealed new information about how cells crank up proteases. The results improve researchers’ understanding of the mechanics of breast cancer and point to where to look for possible indicators of early disease. [Press release from the Pacific Northwest National Laboratory discussing online prepublication in PLoS ONE] Hormone Therapy Use by Postmenopausal Women Associated With Increased Incidence of More Advanced Breast Cancer Follow-up of about 11 years of participants in the Women’s Health Initiative finds that among postmenopausal women, use of estrogen plus progestin is associated with an increased incidence of breast cancers that are more advanced, and with a higher risk of deaths attributable to breast cancer, according to a study. CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal) LABORATORY RESEARCH Cdc42-Interacting Protein 4 Promotes Breast Cancer Cell Invasion and Formation of Invadopodia through Activation of N-WASp Researchers found that expression of CIP4, but not TOCA-1 or FBP17, was increased in invasive breast cancer cell lines in comparison with weakly or noninvasive breast cancer cell lines. [Cancer Res] High Levels of Hsp90 Cochaperone p23 Promote Tumor Progression and Poor Prognosis in Breast Cancer by Increasing Lymph Node Metastases and Drug Resistance To gain a comprehensive view of the effects of p23 on estrogen receptor-dependent and -independent gene expression, researchers profiled mRNA expression from control versus MCF-7+p23 cells in the absence and presence of estrogen. [Cancer Res] Automated Extraction of DNA and RNA From a Single Formalin-Fixed Paraffin-Embedded Tissue Section for Analysis of Both Single-Nucleotide Polymorphisms and mRNA Expression Researchers investigated a fully automated, xylene-free nucleic acid extraction method for the simultaneous analysis of RNA and DNA biomarkers related to breast cancer. [Clin Chem] Genomic Collaboration of Estrogen Receptor-Alpha and ERK2 in Regulating Gene and Proliferation Programs In these studies, researchers document that estrogen-occupied estrogen receptor (ER)-alpha activates and interacts with ERK2, a downstream effector in the MAPK pathway, resulting in ERK2 and ER-alpha colocalization at chromatin binding sites across the genome of breast cancer cells. [Mol Cell Biol] PTP1B Suppresses Prolactin Activation of Stat5 in Breast Cancer Cells Collectively, the data implicate tyrosine phosphatase PTP1B as an important negative regulator of Stat5 phosphorylation in invasive breast cancer. [Am J Pathol] ErbB2 Stabilizes Epidermal Growth Factor Receptor (EGFR) Expression via Erk and Sprouty2 in Extracellular Matrix Detached Cells Researchers show that there is a significant decrease in the stability of EGFR in extracellular matrix-detached cells which is reversed by ErbB2 overexpression. [J Biol Chem] Fetuin-A (ahsg) Is A Major Serum Adhesive Protein That Mediates Growth Signaling in Breast Tumor Cells In the present analyses, researchers show that fetuin-A whose function in cellular attachment in tissue culture has been debated for many years, is indeed a major serum cell attachment factor particularly for tumor cells. [J Biol Chem] A Multigene Predictor of Metastatic Outcome in Early Stage Hormone Receptor-Negative and Triple-Negative Breast Cancer A multigene hormone receptor negative/triple-negative (HRneg/Tneg) signature linked to immune/inflammatory cytokine regulation was identified from pooled expression microarray data and shown to be superior to other reported gene signatures in predicting the metastatic outcome of early stage and conservatively managed HRneg and Tneg breast cancer. [Breast Cancer Res] CLINICAL RESEARCH Estrogen Plus Progestin and Breast Cancer Incidence and Mortality in Postmenopausal Women The objective of this study was to determine the effects of therapy with estrogen plus progestin on cumulative breast cancer incidence and mortality after a total mean follow-up of 11.0 (SD, 2.7) years, through August 14, 2009. [JAMA] Assessment of Clinical Validity of a Breast Cancer Risk Model Combining Genetic and Clinical Information Combining validated common genetic risk factors with clinical risk factors resulted in modest improvement in classification of breast cancer risks in white non-Hispanic postmenopausal women. [J Natl Cancer Inst] Urine Metabolite Analysis Offers Potential Early Diagnosis of Ovarian and Breast Cancers Metabolomics is a new, rapidly expanding field dedicated to the global study of metabolites in biological systems. In this manuscript metabolomics is applied to find urinary biomarkers for breast and ovarian cancer. [Clin Cancer Res] Blood Peptidome-Degradome Profile of Breast Cancer The peptidome-degradome profiles of pooled blood plasma sampled from 15 breast cancer patients and age, race, and menopausal status matched control healthy persons were globally characterized using advanced comprehensive separations combined with tandem Fourier transform mass spectrometry and new data analysis approaches that facilitated top-down peptidomic analysis.
