TOP STORYKv2.1 Clustering Contributes to Insulin Exocytosis and Rescues Human β-Cell Dysfunction Researchers examined the mechanism by which the voltage-dependent K+ (Kv) channel Kv2.1 facilitates depolarization-induced exocytosis in INS 832/13 cells and β-cells from human donors with and without type 2 diabetes. PUBLICATIONS(Ranked by impact factor of the journal)DIABETES & PANCREATITISmTOR Controls ChREBP Transcriptional Activity and Pancreatic β Cell Survival under Diabetic Stress Investigators showed that mice lacking the core nutrient-sensitive signaling component mammalian target of rapamycin (mTOR) in β cells exhibit reduced β cell mass and smaller islets. mTOR deficiency lead to a severe reduction in β cell survival and increased mitochondrial oxidative stress in chemical-induced diabetes. [J Cell Biol] Abstract Interleukin 6 Protects Pancreatic β Cells from Apoptosis by Stimulation of Autophagy Scientists showed that IL-6 robustly activates signal transducer and activator of transcription 3, a transcription factor that is involved in autophagy. IL-6 stimulates LC3 conversion and autophagosome formation in cultured β cells. [FASEB J] Abstract Researchers showed that fluoxetine significantly reduces glucose stimulated insulin secretion. MIN6 cells, an established murine immortalized β cell line, formed smaller colonies of loosely packed cells with reduced cell-cell contact after fluoxetine treatment. [Sci Rep] Full Article The Bile Acid Receptor FXR Attenuates Acinar Cell Autophagy in Chronic Pancreatitis Farnesoid X receptor (FXR) expression, autophagy, apoptosis and necroptosis were determined in human chronic pancreatitis tissue in vivo and in pancreatic cells lines in vitro by means of real-time PCR, immunoblots and immunofluorescence. [Cell Death Discov] Full Article PANCREATIC CANCERScientists examined the functional relationship between cell invasion and TGF-β1 activation in pancreatic ductal adenocarcinoma cells, and confirmed that both processes are αvβ6-dependent. [J Pathol] Abstract Investigators explored the anti-metastatic mechanisms of 8-hydroxydeoxyguanosine (8-OHdG) in Panc-1 pancreatic cancer cells. 8-OHdG inhibited cell migration by inactivating ERM and Rho-GTPase proteins, and inhibiting focal adhesion kinase and matrix metalloproteinases. [Free Radic Biol Med] Abstract Co-Targeting of MEK and PDGFR/STAT3 Pathways to Treat Pancreaticductal Adenocarcinoma Using a two-dimensional monolayer culture system as well as three-dimensional spheroid culture system, scientists conducted a screen of a large panel of anti-cancer agents, and found that MEK inhibitors were most effective in targeting pancreatic ductal adenocarcinoma spheroids in comparison to monolayer cultures. [Mol Cancer Ther] Abstract The authors demonstrated a significant association between C1QBP expression and hepatic metastasis in patients with pancreatic cancer. Insulin-like growth factor-1 (IGF-1) induced the translocation of C1QBP from cytoplasm to lipid rafts and further drove the formation of CD44 variant 6/C1QBP complex in pancreatic cancer cells. [Int J Cancer] Abstract Investigators demonstrated the underlying mechanism of miR-148a in the regulation of epithelial-mesenchymal transition and invasion of pancreatic cancer cells. They revealed that the expression of miR-148a was markedly downregulated in human pancreatic ductal adenocarcinoma cell lines and tissues. [Oncol Rep] Abstract | |

POLICY NEWSChina Cracks Down on Fake Peer Reviews The Chinese government is going on the offensive against scientists who dupe journals by creating fraudulent reviews of submitted papers. A coalition of agencies led by the science ministry announced that the government would suspend the grants of researchers involved in such fraud, which surfaced earlier this year when a cancer journal retracted 107 research papers from Chinese authors. [Nature News] Editorial Strengthened US Sanctions on Cuba Disappoint Scientists US President Donald Trump announced that he would strengthen travel and trade restrictions against Cuba, reversing his predecessor’s attempts to normalize relations with the country. The move is a blow to scientists who hoped that former president Barack Obama’s push to relax US restrictions on Cuba would make it easier for researchers there to travel and to collaborate with colleagues in the United States. [Nature News] Editorial Macron Consolidates Electoral Victory The party of France’s recently elected president won an absolute majority in its first general elections, with an agenda that included strong support for research.
