TOP STORYMNK1 Signaling Induces an ANGPTL4-Mediated Gene Signature to Drive Melanoma Progression Researchers showed that changes in the activity, or abundance, of MAP kinase-interacting serine/threonine-protein kinase 1 (MNK1) resulted in changes in the expression of pro-oncogenic and pro-invasive genes. Among the MNK1-upregulated genes, they identified angiopoietin-like 4 (ANGPTL4), which in turn promoted an invasive phenotype via its ability to induce the expression of matrix metalloproteinases. PUBLICATIONS(Ranked by impact factor of the journal)DERMAL STEM CELLS & TISSUE REGENERATIONHistone Variant Dictates Fate Biasing of Neural Crest Cells to Melanocyte Lineage Scientists established a role of a histone variant H2a.z.2 in specification of melanocyte lineage from multipotent neural crest cells. Silencing of H2a.z.2 reduced the number of melanocyte precursors in developing zebrafish embryos, and from mouse embryonic stem cells in vitro. [Development] Abstract Functional Consequences of PDK4 Deficiency in Doberman Pinscher Fibroblasts Investigators explored the functional consequences of pyruvate dehydrogenase kinase 4 (PDK4) deficiency in Doberman Pincher fibroblasts representing PDK4wt/wt, PDK4wt/del, and PDK4del/del genotypes. [Sci Rep] Full Article Researchers examined the outcome of infection of cultured human keratinocyte cells with La Crosse virus, using a spontaneously transformed cell line, HaCaT. [Viruses] Abstract | Download Full Article Accessing the Transcriptional Status of Selenoproteins in Skin Cancer-Derived Cell Lines Using RNA-seq data set from Sequence Reads Archive experiments published at the National Center for Biotechnology Information, investigators determined and compared the transcriptional levels of the 25 selenoproteins-coding sequences found in 16 human-derived melanoma cell lines and compared to four melanocyte controls. [J Trace Elem Med Biol] Abstract | Graphical Abstract The authors investigated whether infrared‐A (IRA) inhibits the proliferation of normal human epidermal keratinocytes (NHEKs). IRA irradiation resulted in cell cycle arrest at G1 and a dose‐dependent reduction in the proliferation of NHEKs. [Photochem Photobiol] Abstract Scientists evaluated the effects of commercialized denture adhesives and mouth moisturizers using direct and indirect cell cultures for in vitro examinations with human fibroblast and epithelial cells. [Dent Mater J] Abstract Investigators adapted a previously published full-thickness skin- equivalent (SE) model by seeding increasing ratios of stress-induced premature senescent versus normal fibroblasts into the collagen matrix, terming these SE “senoskin”. [NPJ Aging Mech Dis] Full Article SKIN CANCERS & DISORDERSThe authors studied the effects of β3-tubulin gene silencing on microtubule (MT) dynamics, cell cycle, and microvesicle (MV) release in human malignant melanoma cells. The knockdown of β3-tubulin induced G2/M cell cycle arrest, impaired MT dynamics, and reduced spontaneous MV release. [Int J Mol Sci] Full Article Researchers showed that ethanol extracts of Angelica dahurica Radix suppressed cell growth and induced apoptotic cell death in B16F10 cells. REVIEWSAn Update on the Implications of Cyclin D1 in Melanomas The authors examine data published on upregulation of the CCND1 gene and cyclin D1 protein in the melanoma setting, focusing on the pathways and molecular mechanisms involved in the activation of the gene and on the clinical and therapeutic implications. [Pigment Cell Melanoma Res] Abstract Mitochondrial Sirtuins in Skin and Skin Cancers Scientists discuss the roles and implications of mitochondrial sirtuins in cutaneous cellular processes, and their emerging potential as a target for the management of skin diseases, including skin cancer.
