Pak1 Maintains Epidermal Stem Cells by Regulating Langerhans Cells and Is Required for Skin Carcinogenesis

The authors suggested that Pak1 regulated the epidermal stem cell number by changing the properties of Langerhans cells (LCs) and functions in skin carcinogenesis. MSC-Derived Exosomes Attenuate Cell Death through Suppressing AIF Nucleus Translocation and Enhance Cutaneous Wound Healing

Scientists found that human umbilical cord mesenchymal stem cells (MSCs) treatment significantly increased immortalized human keratinocytes (HaCaT) cell proliferation and migration in a time- and dose-dependent manner, suppressed HaCaT apoptosis induced with H₂O₂ by inhibiting nuclear translocation of apoptosis-inducing factor. Vanillic Acid Stimulates Anagen Signaling via the PI3K/Akt/β-Catenin Pathway in Dermal Papilla Cells

The authors showed that vanillic acid, a phenolic acid from wheat bran, promoted the proliferation of dermal papilla cells (DPCs) via a PI3K/Akt/Wnt/β-catenin dependent mechanism. Vanillic acid promoted the proliferation of DPCs, accompanied by increased levels of cell-cycle proteins cyclin D1, CDK6, and Cdc2 p34. [Biomol Ther] Abstract

MiR-23b Promotes the Migration of Keratinocytes through Downregulating TIMP3

Investigators showed that tissue inhibitor of metalloproteinase-3 (TIMP3) was a direct target of miR-23b and was negatively regulated by miR-23b. TIMP3 inhibited the migration of keratinocytes. MiR-23b accelerated the migration of keratinocytes by downregulating the abundance of TIMP3. [J Surg Res] Abstract

Integrin α3β1 in Hair Bulge Stem Cells Modulates CCN2 Expression and Promotes Skin Tumorigenesis

Using a reporter mouse line with inducible deletion of α3β1 in hair bulge stem cells (HB SCs), researchers showed that HB SCs remained confined to their niche regardless of the presence of α3β1 and were largely absent from skin tumors. However, tumor formation was significantly decreased in mice deficient for α3β1 in HB SCs. [Life Sci Alliance] Full Article

Triterpenoid Acids Isolated from Schinus terebinthifolia Fruits Reduce Staphylococcus aureus Virulence and Abate Dermonecrosis

Investigators reported the isolation and identification of three compounds from 430D-F5, a refined Schinus terebinthifolia fruits extract, that reduced virulence and abate dermonecrosis. Each compound inhibited all S. aureus accessory gene regulator alleles. [Sci Rep] Full Article

Differential Responsiveness to BRAF Inhibitors of Melanoma Cell Lines BRAF^V600E-Mutated

The authors tested whether three cell lines, discordant in the mutation detection, responded differently to BRAF-specific inhibitors. Their proliferation rate decreased after treatment with vemurafenib and PLX4720 but was not affected by sorafenib, suggesting a BRAF^V600E biological behavior. [J Transl Med] Full Article

Sorafenib Induces Pigmentation via the Regulation of β-Catenin Signaling Pathway in Melanoma Cells

Scientists investigated the effect of sorafenib on the intracellular signaling pathways. Sorafenib reduced phosphorylation of AKT and ERK, suggesting that sorafenib induced pigmentation through inhibition of the AKT and ERK pathways. [Exp Dermatol] Abstract

Role of SPRED1 in Keratinocyte Proliferation in Psoriasis

To further investigate the molecular mechanism by which psoriatic dermal mesenchymal stem cells stimulate keratinocyte proliferation, and the role of missense SPRED1 mutations in psoriasis, researchers assessed expression levels of miRNA, and both mRNA and protein of SPRED1 in normal human epidermal keratinocyte cells.
