The Glucose Transporter GLUT3 Controls T Helper 17 Cell Responses through Glycolytic-Epigenetic Reprogramming

Scientists showed that glycolytic reprogramming additionally controlled inflammatory gene expression via epigenetic remodeling. They found that the glucose transporter GLUT3 was essential for the effector functions of Th17 cells in models of autoimmune colitis and encephalomyelitis.

Pirfenidone Exacerbates Th2-Driven Vasculopathy in a Mouse Model of SSc-ILD

Pirfenidone treatment attenuated pulmonary remodeling in the bleomycin-model, but aggravated pulmonary inflammation, fibrosis, and vascular remodeling in Fra-2 overexpressing mice.

Cell Death-Induced Immunogenicity Enhances Chemoimmunotherapeutic Response by Converting Immune-Excluded Into T Cell Inflamed Bladder Tumors

Researchers explored an alternative, drug-induced mechanism that impedes therapeutic response via disrupting the onset of immunogenic cell death.

Cell-Free Extracts from Human Fat Tissue with a Hyaluronan-Based Hydrogel Attenuate Inflammation in a Spinal Cord Injury Model through M2 Microglia/Microphage Polarization

Fat extract (FE) is a cell-free liquid component containing a variety of cytokines extracted from human fat tissue using mechanical methods. A biocompatible hyaluronan and methylcellulose loaded with FE is used to treat a model of spinal cord contusion in mice.

A Peptide Encoded by Pri-miRNA-31 Represses Autoimmunity by Promoting Treg Differentiation

miPEP31 was highly expressed in Foxp3+ Tregs and significantly promoted the differentiation of Tregs without affecting their inhibitory ability.

HIPK2 Directs Cell Type–Specific Regulation of STAT3 Transcriptional Activity in Th17 Cell Differentiation

The authors reported that in mouse T helper 17 (Th17) cells, signal transducer and activator of transcription 3 (Stat3) phosphorylation at serine 727, required for its transcriptional activity, was carried out by homeodomain-interacting protein kinase 2 (HIPK2), a nuclear kinase selectively up-regulated in Th17 cells.

    Abstract

CSF-1 Maintains Pathogenic but Not Homeostatic Myeloid Cells in the Central Nervous System during Autoimmune Neuroinflammation

Investigators found that targeting colony stimulating factor 1 (CSF-1) with neutralizing antibody halted ongoing encephalomyelitis, with efficacy superior to colony CSF-1 receptor inhibitor BLZ945, whereas IL-34 neutralization had no effect, suggesting that pathogenic myeloid cells were maintained by CSF-1.

Carbon Ion Radiotherapy Triggers Immunogenic Cell Death and Sensitizes Melanoma to Anti-PD-1 Therapy in Mice

Scientists used human U2OS osteosarcoma cells for the in vitro assessment of immunogenic cell death and established several in vivo models of melanoma in mice.

Alpha-7 Nicotinic Acetylcholine Receptor Agonist Alleviates Psoriasis-Like Inflammation through Inhibition of the STAT3 and NF-κB Signaling Pathway

Researchers analyzed the expression of alpha-7 nicotinic acetylcholine receptor (α7nAchR )in human and mouse psoriatic skin tissue. In vivo, PNU-282987 or Methyllycaconitine, a specific agonist or antagonist of α7nAchR, were administered to imiquimod-induced psoriatic mouse models.

The Pancreatic Cancer Immune Tumor Microenvironment Is Negatively Remodeled by Gemcitabine While TGF-β Receptor plus Dual Checkpoint Inhibition Maintains Antitumor Immune Cells

The authors hypothesized that TGF-β receptor inhibition may alter pancreatic ductal adenocarcinoma progression and antitumor immunity in the tumor microenvironment.

Roles of RNA-Binding Proteins in Immune Diseases and Cancer

Investigators summarize the recent progress in our understanding of the functional roles of RNA-binding proteins (RBPs) in several types of immune responses, and the involvement of RBP dysregulation in the pathogenesis of immune diseases and cancer.
