Evolutionary Differences in the VEGF/VEGFR Code Reveal Organotypic Roles for the Endothelial Cell Receptor KDR in Developmental Lymphangiogenesis

Scientists defined the zebrafish VEGF/VEGFR code in receptor binding studies. They found that while VEGFD directed craniofacial lymphangiogenesis, it bound KDR but surprisingly, unlike in mammals, did not bind FLT4. [Cell Rep] Full Article | Graphical Abstract

Endothelial-to-Mesenchymal Transition in Human Adipose Tissue Vasculature Alters the Particulate Secretome and Induces Endothelial Dysfunction

Researchers showed that adipose tissue endothelial cells exposed in vitro to tumor growth factor-β, tumor necrosis factor-α, and interferon-γ underwent endothelial-to-mesenchymal transition with progressive loss of endothelial markers. YBX-1 Mediated Sorting of miR-133 into Hypoxia/Reoxygenation-Induced EPC-Derived Exosomes to Increase Fibroblast Angiogenesis and MEndoT

Hypoxia/reoxygenation treatment promoted miR-133 expression in endothelial progenitor cells (EPCs) and EPC-derived exosomes. miR-133 was incorporated into exosomes and transmitted to cardiac fibroblasts, increasing the angiogenesis and mesenchymal-endothelial transition (MEndoT) of cardiac fibroblasts. TRPM7 Mediates Kidney Injury, Endothelial Hyperpermeability and Mortality during Endotoxemia

The authors showed that endotoxin increases endothelial hyperpermeability and Ca²⁺ overload through the TLR4/NOX-2/ROS/NF-κB pathway. Moreover, endotoxin exposure was shown to downregulate the expression of VE-cadherin, compromising monolayer integrity and enhancing vascular hyperpermeability. [Lab Invest] Abstract

In CDh5-CreERT2VEGFR2lox/PGFD mice, conditional deletion of VEGF receptor 2 in vascular endothelial cells abolished injury-induced hemangiogenesis and lymphangiogenesis, demonstrating the utility of this transgenic mouse line for identifying important factors in the process of neovascularization. Stachydrine Mediates Rapid Vascular Relaxation: Activation of Endothelial Nitric Oxide Synthase Involving AMP-Activated Protein Kinase and Akt Phosphorylation in Vascular Endothelial Cells

Stachydrine (STA) activated AMP-activated protein kinase (AMPK), protein kinase B/Akt, and endothelial nitric oxide synthase (eNOS) in vascular endothelial cells. Endothelial TGF-β Signaling Drives Vascular Inflammation and Atherosclerosis

Researchers showed that endothelial transforming growh factor β (TGF-β) signaling was one of the primary drivers of atherosclerosis-associated vascular inflammation. Inhibition of endothelial TGF-β signaling in hyperlipidemic mice reduced vessel wall inflammation and vascular permeability and lead to arrest of disease progression and regression of established lesions.
