Single-Cell RNA Sequencing Maps Endothelial Metabolic Plasticity in Pathological Angiogenesis

By single-cell RNA sequencing 28,337 murine choroidal ECs (CECs) and sprouting horoidal neovascularization-ECs, researchers constructed a taxonomy to characterize their heterogeneity. Comparison with murine lung tumor ECs revealed congruent marker gene expression by distinct EC phenotypes across tissues and diseases, suggesting similar angiogenic mechanisms. ADAMTS-1 and Syndecan-4 Intersect in the Regulation of Cell Migration and Angiogenesis

Knockdown of ADAMTS-1 in endothelial cells resulted in a parallel reduction in cell surface syndecan-4, attributable to increased matrix metalloproteinase-9 activity. Knockdown of either ADAMTS-1 or syndecan-4 increased cellular responses to vascular endothelial growth factor A isoform VEGFA₁₆₄, and increased ex vivo aortic ring microvessel sprouting. Fasentin Diminishes Endothelial Cell Proliferation, Differentiation and Invasion in a Glucose Metabolism-Independent Manner

Scientists tested the capability of fasentin to modulate angiogenesis in vitro and in vivo. They showed that fasentin inhibited tube formation in endothelial cells by a mechanism that involved a negative effect on endothelial cell proliferation and invasion, without affecting other steps related to the angiogenic process. The Epigallocatechin Gallate Derivative Y6 Inhibits Human Hepatocellular Carcinoma by Inhibiting Angiogenesis in MAPK/ERK1/2 and PI3K/AKT/HIF-1α/VEGF Dependent Pathways

The authors evaluated the effects of epigallocatechin gallate, Y6 on hepatocellular carcinoma and angiogenesis in vivo and in vitro. Injectable Therapeutic Organoids Using Sacrificial Hydrogels

Investigators introduced a method of using hydrogels as sacrificial scaffolds, which allowed cells to form self-organized clusters followed by gentle release, resulting in highly reproducible multicellular structures on a large scale. They demonstrated this strategy for endothelial cells and mesenchymal stem cells to self-organize into blood-vessel units, which were injected into mice, and rapidly formed perfusing vasculature.
