CRISPR Screening Uncovers a Central Requirement for HHEX in Pancreatic Lineage Commitment and Plasticity Restriction

HHEX cooperated with the pioneer transcription factors (TFs) FOXA1, FOXA2 and GATA4, shared by both pancreas and liver differentiation programmes, to promote pancreas commitment, and this cooperation restrained the shared TFs from activating alternative lineages.

    Abstract

Performance of Islets of Langerhans Conformally Coated via an Emulsion Cross-Linking Method in Diabetic Rodents and Nonhuman Primates

The authors developed and validated an augmented conformal coating (CC) method that achieved hydrogel CCs around islets at physiological pH for improved cytocompatibility, with polyethylene glycol hydrogels for increased biocompatibility and with fivefold increase in encapsulation throughput for enhanced scalability.

Proinflammatory Signaling in Islet β Cells Propagates Invasion of Pathogenic Immune Cells in Autoimmune Diabetes

Scientists deleted the proinflammatory gene encoding 12/15-lipoxygenase in β cells of non-obese diabetic mice at a pre-diabetic time point when islet inflammation was a feature.

Coordinated Transcriptional and Catabolic Programs Support Iron Dependent Adaptation to RAS-MAPK Pathway Inhibition in Pancreatic Cancer

Using transcriptome and chromatin immunoprecipitation profiling of PDAC cells treated with the MEK inhibitor, Trametinib, researchers identified transcriptional antagonism between c-MYC and the master transcription factors for lysosome gene expression, the MiT/TFE proteins.

    Abstract

Acetyl-coA Synthetase 2 Potentiates Macropinocytosis and Muscle Wasting through Metabolic Reprogramming in Pancreatic Cancer

Metabolic stress induced ACSS2 expression, which is associated with worse prognosis in pancreatic cancer. ACSS2 knockout significantly suppressed cell proliferation in 2D and 3D models.

MARK2 Regulates Chemotherapeutic Responses Through Class IIa HDAC-YAP Axis in Pancreatic Cancer

Through a Phos-tag-based kinome-wide screen, the authors identified microtubule affinity-regulating kinase proteins (MARK2) as a critical regulator for paclitaxel chemosensitivity in PDAC.

Riluzole Regulates Pancreatic Cancer Cell Metabolism by Suppressing the Wnt-β-Catenin Pathway

Investigators reported that riluzole inhibited pancreatic cancer cell growth and had no effect on human pancreatic normal ductal epithelial cells.

N(6)-Methyladenosine Methylation-Regulated Polo-Like Kinase 1 Cell Cycle Homeostasis as a Potential Target of Radiotherapy in Pancreatic Adenocarcinoma

By epitranscriptomic analysis, researchers reported that polo-like kinase 1 was an important hub gene defining patient prognosis in pancreatic cancer and that RNA methylation was involved in regulating its cell cycle-specific expression.
