Periostin Activation of Integrin Receptors on Sensory Neurons Induces Allergic Itch

Using pharmacological and genetic approaches, scientists inhibited the function of neuronal integrin αVβ3, which significantly reduced periostin-induced itch in mice. Autophosphorylation at Serine 166 Regulates RIP Kinase 1-Mediated Cell Death and Inflammation

The authors showed that serine 166 (S166) autophosphorylation licensed receptor interacting protein kinase (RIPK1) activity to induce downstream cell death signaling and inflammation, suggesting that S166 phosphorylation could serve as a reliable biomarker for RIPK1 kinase-dependent pathologies. Researchers demonstrated that T-LAK cell-originated protein kinase (TOPK) could phosphorylate and activate c-Jun at Ser63 and Ser73 in the process of skin carcinogenesis and HI-TOPK-032 could be used as a potential chemopreventive drug against cutaneous squamous cell carcinoma development. TRAF6 Activates Fibroblasts to Cancer-Associated Fibroblasts (CAFs) through FGF19 in Tumor Microenvironment to Benefit the Malignant Phenotype of Melanoma Cells

Investigators found that TRAF6 was significantly upregulated in CAFs adjacent to melanoma cells. They demonstrated that TRAF6 promoted fibroblast proliferation and migration as well as MMP and α-SMA expression. TRAF6 expression in fibroblasts promoted the malignant phenotype of melanoma cells in vitro and in vivo. [J Invest Dermatol] Abstract

Autophagy Inhibition Potentiates the Anti-EMT Effects of Alteronol through TGF-β/Smad3 Signaling in Melanoma Cells

Scientists suggested that alteronol induced cyto-protective autophagy in melanoma cells through inhibition of Akt/mTOR pathway, attenuating apoptosis and promoting melanoma cell epithelial-mesenchymal transition (EMT) through TGF-β/Smad3 pathway. [Cell Death Dis] Full Article

Collagen-Binding IL-12 Enhances Tumor Inflammation and Drives the Complete Remission of Established Immunologically Cold Mouse Tumors

Researchers found that CBD–IL-12 potently synergized with checkpoint-inhibitor to eradicate large established melanomas, induced antigen-specific immunological memory and controlled tumor growth in a genetically engineered mouse model of melanoma. [Nat Biomed Eng] Abstract

Identification of Very Small Cancer Stem Cells Expressing Hallmarks of Pluripotency in B16F10 Melanoma Cells and Their Reoccurrence in B16F10-Derived Clones

The authors identified rare cells in the murine melanoma B16F10, and human A2058 and SK-MEL-28 cell lines that expressed pluripotency markers, including Oct4, Nanog, Sox2 and a marker of melanoma cancer cells. [Exp Cell Res] Abstract
