KRAS(G12D) Drives Lepidic Adenocarcinoma through Stem-Cell Reprogramming

Scientists demonstrated that expression of KRAS(G12D) in differentiated alveolar epithelial type I (AT1) cells reprogrammed them slowly and asynchronously back into AT2 stem cells that go on to generate indolent tumors.

Glucose Transporter 1-Mediated Transcytosis of Glucosamine-Labeled Liposomal Ceramide Targets Hypoxia Niches and Cancer Stem Cells to Enhance Therapeutic Efficacy

Experimental results indicated that glucosamine-labeled liposomal ceramide could be efficiently transported between cancer cells by GLUT1 transporters and substantially accumulated in the hypoxic area in in vitro CSC spheroids and in vivo tumor xenografts.

circSLC4A7 Accelerates Stemness and Progression of Gastric Cancer by Interacting with HSP90 to Activate NOTCH1 Signaling Pathway

Researchers discovered the expression profile and functional roles of circRNAs in gastric CSCs (GCSCs). A spheroid formation assay was conducted to enrich GCSCs. Genome-wide sequencing of circRNAs showed that a novel circRNA, circSLC4A7, was one of the most upregulated circRNAs in GCSCs.

Deciphering Brain Metastasis Stem Cell Properties from Colorectal Cancer Highlights Specific Stemness Signature and Shared Molecular Features

Four brain metastasis stem cell lines from patients with colorectal cancer were obtained by mechanical dissociation of patient’s tumors and selection of CSCs by appropriate culture conditions.

Chimeric Antigen Receptor T Cells Targeting Cell Surface GRP78 Efficiently Kill Glioblastoma and Cancer Stem Cells

To target the cell surface glucose-regulated protein 78 (GRP78), CAR-T cells based on its binding peptide were generated. In vitro two glioblastoma cell lines and glioma stem cells were used to confirm the localization of cell surface GRP78 and the cytotoxicity of the CAR-T cells.

B7-H3 Confers Stemness Characteristics to Gastric Cancer Cells by Promoting Glutathione Metabolism through AKT/pAKT/Nrf2 Pathway

Gastric cancer stemness influenced by B7-H3 was detected both in vitro and in vivo. The expression of stemness-related markers was examined by reverse transcriptase quantitative polymerase chain reaction, Western blotting, and flow cytometry.

Propofol Inhibits Colon Cancer Cell Stemness and Epithelial-Mesenchymal Transition by Regulating SIRT1, Wnt/β-Catenin, and PI3K/AKT/mTOR Signaling Pathways

The authors used propofol to treat LOVO and SW480 cells and Cell Counting Kit-8 to detect proliferation. Self-renewal capacity, cell invasion and migration, flow cytometry analysis, qPCR and Western blotting were performed to detect the suppression of propofol to colon cancer cells and the underlying mechanism.

KRAS Activation in Gastric Cancer Stem-Like Cells Promotes Tumor Angiogenesis and Metastasis

Scientists analyzed how KRAS activation in gastric CSCs promoted tumor angiogenesis and metastasis. Gastric cancer CSCs were found to secrete pro-angiogenic factors such as vascular endothelial growth factor A, and inhibition of KRAS markedly reduced secretion of these factors.

Anisomycin Inhibits the Activity of Human Ovarian Cancer Stem Cells via Regulating Antisense RNA NCPB2-AS2/MEK/ERK/STAT3 Signaling

CD44+/CD133+ human ovarian cancer stem cells (OCSCs) were isolated from human ovarian cancer tissues. OCSCs were interfered with using anisomycin and specific small-interfering RNA.

Ferroptosis as a Potential Target for Cancer Therapy

Investigators summarize and briefly present the key pathways of ferroptosis, paying special attention to the regulation of ferroptosis as well as its dual role as an oncogenic and as a tumor suppressor event in various human cancers.
