    Identification of Small Molecules that Support Human Leukemia Stem Cell Activity Ex Vivo
    Currently available culture conditions do not prevent spontaneous differentiation of leukemic stem cells (LSCs), which greatly limits the feasibility of cell-based assays. To overcome these constraints investigators conducted a high-throughput chemical screen and identified small molecules that inhibit differentiation and support LSC activity in vitro.     Scientists showed a reduction of asymmetrical cell division in spheroid-derived colorectal cancer stem cells compared with differentiated cancer cells. [Nat Cell Biol] Abstract

    Analysis of the Tumor-Initiating and Metastatic Capacity of PDX1-Positive Cells from the Adult Pancreas
    The authors sought to determine which subsets of pancreatic and duodenal homeobox 1 (PDX1)+ cells may be responsible for tumor growth. Using the Lox-Stop-Lox-KrasG12D genetic mouse model of pancreatic carcinogenesis, they isolated a population of KrasG12D-expressing PDX1+ cells with an inherent capacity to metastasize.     Effect of Niclosamide on Basal-Like Breast Cancers
    Aldehyde dehydrogenase (ALDH) is a known marker of cancer stem cells. By testing basal-like breast cancer cells for ALDH expression by flow cytometry, investigators were able to isolate a non-adherent population of cells that have high ALDH expression. [Mol Cancer Ther] Abstract

    Patients with CD133-Negative Colorectal Liver Metastasis Have a Poor Prognosis after Hepatectomy
    Resected specimens were obtained surgically from 1998 to 2008 at a university hospital. Researchers investigated whether the status of two primary lesion cancer stem biomarkers, CD44 and CD133, were maintained in liver metastasis (L-Mets) and whether these markers were L-Mets prognostic factors.     ER-α36-Mediated Rapid Estrogen Signaling Positively Regulates ER-Positive Breast Cancer Stem/Progenitor Cells
    Researchers investigated the function and the underlying mechanism of 36 kDa full-length estrogen receptor-α (ER-α36)-mediated rapid estrogen signaling in growth regulation of the ER-positive breast cancer stem/progenitor cells. [PLoS One] Full Article

    USP22 Is a Positive Regulator of NFATc2 on Promoting IL2 Expression
    Investigators showed that ubiquitin-specific protease 22 (USP22), known as a cancer stem cell marker, specifically interacted with and deubiquitinated nuclear factor of activated T cells, cytoplasmic 2 (NFATc2). [FEBS Lett] Abstract

    The NFκB Inhibitor, SN50, Induces Differentiation of Glioma Stem Cells and Suppresses Their Oncogenic Phenotype
    Scientists show that SN50, a cell-permeable peptide inhibitor of NFκB, induced robust differentiation of human glioma stem-like cells, causing loss of their oncogenic potential. [Cancer Biol Ther] Full Article

    Knockdown of 14-3-3ζ Enhances Radiosensitivity and Radio-Induced Apoptosis in CD133+ Liver Cancer Stem Cells
    Researchers investigated whether silencing 14-3-3ζ affects the radioresistance of cancer stem-like cells (CSCs) in hepatocellular carcinoma. Knockdown of 14-3-3ζ decreased cell viability and the number of spheres by reducing radioresistance in CSCs after γ-irradiation. [Exp Mol Med] Full Article
