TOP STORYTargeting SRC Coactivators Blocks the Tumor-Initiating Capacity of Cancer Stem-Like Cells Investigators report that steroid receptor coactivator (SRC)-3 supports the tumor initiating cell/CSC state and induces an epithelial to mesenchymal transition (EMT) by driving expression of the master EMT regulators and stem cell markers. PUBLICATIONS(Ranked by impact factor of the journal)Downregulation of PD-L1 compromised the self-renewal capability of breast CSCs in vitro and in vivo as shown by tumorsphere formation assay and extreme limiting dilution assay. [Int J Cancer] Abstract Targeting Glioma Stem Cells In Vivo by a G-Quadruplex-Stabilizing Synthetic Macrocyclic Hexaoxazole Researchers demonstrated that a synthetic G4 ligand, Y2H2-6M(4)-oxazole telomestatin derivative, limits the growth of intractable glioblastoma and glioma stem cells. [Sci Rep] Full Article CFC1 Is a Cancer Stemness-Regulating Factor in Neuroblastoma The overexpression of CFC1 increased sphere formation, cell growth, and colony formation. These phenotypes, particularly sphere formation, and xenograft tumor formation were significantly suppressed by the knockdown of CFC1. [Oncotarget] Full Article As single agents, curcumin and metformin, are reported to exhibit chemopreventive properties, in vitro as well as in patients with oral cancer. The chemopreventive efficacy of this drug combination was tested in a 4-nitro-quinoline-oxide induced, mice oral carcinogenesis model. [Mol Carcinog] Abstract Stable SLC7A11 knock-down and over-expressing U251 glioma cells were generated and characterized to understand the role of redox and system xc– in glioma progression. SLC7A11 knock-down resulted in higher endogenous reactive oxygen species levels and enhanced invasive properties. [Stem Cells Dev] Abstract The authors investigated the mechanism of the oncogenic effects of human growth hormone (hGH) in hepatocellular carcinoma (HCC) cell lines. In vitro functional assays demonstrated that forced expression of hGH in these HCC cell lines promoted cell proliferation, cell survival, anchorage-independent growth, cell migration, and invasion, as previously reported. [Int J Mol Sci] Abstract Hm2E8b-NCTD-liposomes selectively ablated B-lineage leukemia stem cells (LSCs) through targeting hepatic leukemia factor (HLF), which is implicated in hematopoietic stem cell regulation and is overexpressed in LSCs. [J Drug Target] Abstract Nanotechniques Inactivate Cancer Stem Cells Scientists present data on phenotype characteristics of Ehrlich carcinoma cells as a convenient and easy-to-follow model of tumor growth. The evidence of cancer stem cells as a part of Ehrlich carcinoma and significance of CD44+ and CD44– subpopulations in maintaining the growth of this type of tumor were demonstrated. [Nanoscale Res Lett] Full Article Zileuton did not exert significant overall growth inhibition in the long-term culture-initiating cell and colony assays of bone marrow CD34+CD38− cells from seven chronic myeloid leukemia patients. The majority of the single leukemic BCR-ABL+CD34+CD38− cells expressed cysteinyl leukotriene receptors CYSLT1 and CYSLT2. [Biochem Biophys Res Commun] Abstract | |

REVIEWSCurcumin and modified derivatives have been shown to have potent anti-cancer effects, especially on CSCs. Berberine is also isolated from various plants and has been used for centuries in traditional medicine to treat diseases such as adult-onset diabetes. Understanding the benefits of these and other nutraceuticals may result in approaches to improve human health. POLICY NEWSUniversity of Tokyo Scientist Hit by Anonymous Allegations Fights Back Anonymous allegations of questionable data and images in 22 papers by six prominent groups at the prestigious University of Tokyo prompted the school to set up an investigating committee. Now, even before the panel completes its investigation, one of the accused researchers has mounted a staunch defense of his work, [ScienceInsider] Editorial China Cracks Down on Fake Peer Reviews The Chinese government is going on the offensive against scientists who dupe journals by creating fraudulent reviews of submitted papers. A coalition of agencies led by the science ministry announced that the government would suspend the grants of researchers involved in such fraud, which surfaced earlier this year when a cancer journal retracted 107 research papers from Chinese authors. [Nature News] Editorial Strengthened US Sanctions on Cuba Disappoint Scientists US President Donald Trump announced that he would strengthen travel and trade restrictions against Cuba, reversing his predecessor’s attempts to normalize relations with the country. The move is a blow to scientists who hoped that former president Barack Obama’s push to relax US restrictions on Cuba would make it easier for researchers there to travel and to collaborate with colleagues in the United States.
