Vasculogenesis in Experimental Stroke after Human Cerebral Endothelial Cell Transplantation
Adult rats were exposed to transient occlusion of the middle cerebral artery and three hours later randomly stereotaxically transplanted with 100K, 200K, or 400K human cerebral endothelial cell six viable cells or vehicle. Animals underwent neurological examination and motor test up to day seven after transplantation then euthanized for immunostaining against neuronal, vascular, and specific human antigens. By utilizing two functionally distinct human mesenchymal stem cell (MSC) clones, researchers found that so-called “pericytic” MSCs secrete the pro-angiogenic vascular guidance molecule SLIT3, which guides vascular development by directing ROBO4-positive endothelial cells to form networks in engineered tissue.
