Suppression of Cytokine Release Syndrome During CAR-T-Cell Therapy via a Subcutaneously Injected Interleukin-6-Adsorbing Hydrogel

Researchers showed that a temperature-sensitive hydrogel conjugated with antibodies for the pro-inflammatory cytokine interleukin-6 (IL-6) and subcutaneously injected before the infusion of CAR-T cells substantially reduces the levels of IL-6 during CRS while maintaining the therapy’s antitumor efficacy.

Alternative Splicing of Its 5′ UTR Limits CD20 mRNA Translation and Enables Resistance to CD20-Directed Immunotherapies

Investigators showed that the MS4A1 gene encoding human CD20 also produces several mRNA isoforms with distinct 5′ untranslated regions. Four variants were detectable by RNA-seq in distinct stages of normal B cell differentiation and B lymphoid malignancies, with V1 and V3 being the most abundant by far.

The Anti-CD40L Monoclonal Antibody AT-1501 Promotes Islet and Kidney Allograft Survival and Function in Nonhuman Primates

An anti-CD40L-specific monoclonal antibody, AT-1501 (Tegoprubart), was engineered to minimize risk of thromboembolic complications by reducing binding to Fcγ receptors expressed on platelets while preserving binding to CD40L.

    Pre-Clinical Validation of a Pan-Cancer CAR-T Cell Immunotherapy Targeting nfP2X7

Investigators presented a prototype -human CAR-T construct targeting nfP2X7 showing potential antigen-specific cytotoxicity against 12 solid cancer types. In xenograft mouse models of breast and prostate cancer, CAR-T cells targeting nfP2X7 exhibit robust anti-tumor efficacy.

ERK1-Mediated Immunomodulation of Mesenchymal Stem Cells Ameliorates Inflammatory Disorders

The authors indicated that ERK1-/- BMMSCs, but not ERK2-/- bone marrow MSCs, failed to promote T cell apoptosis due to incapacity to activate the ETS2/AURKA/NFκB/Fas/MCP-1 cascade.

Umbilical Cord Mesenchymal Stem Cells Relieve Osteoarthritis in Rats Through Immunoregulation and Inhibition of Chondrocyte Apoptosis

In vitro experiments showed that co-culturing umbilical cord MSCs and chondrocytes promoted the expression of aggrecan, COL2, SOX9, and BCL-2; downregulated the expression of BAX and BAD in chondrocytes; and promoted the expression of IL-10 and TGF-β1 in UCMSCs.

Stem Cell Therapy for Degenerative Disc Disease: Bridging the Gap Between Preclinical Promise and Clinical Potential

Stem cell therapy still faces multiple challenges, such as the durability and survival of mesenchymal stem cells upon implantation, uncertain pathways to discogenic differentiation, and the adverse impact of a harsh microenvironment on cell survival.
