    Co-Administration of Human MSC Overexpressing HIF-1α Increases Human CD34+ Cell Engraftment In Vivo

    Human cord blood CD34+ cells were co-cultured with mesenchymal stromal cells (MSC) wild type or hypoxia-inducible factor-1α-MSC (4:1) for 72 hours. Viability, cell cycle, ROS expression and immunophenotyping of key molecules involved in engraftment were evaluated by flow cytometry in CD34+ cells.

    Scientists revealed that the levels of 5-methylcytosine (5mC), were significantly increased coupled with decreased activity of Ten-Eleven Translocation (TETs) enzymes that principally functioned by oxidizing 5mC in the obese placenta, consistent with hypoxia-induced genome-wide DNA hypermethylation observed in varied types of cells and tissues.
