    TOP STORY
    Skin Cells Play ‘Dice Games’
Scientists showed that all dividing skin cells can flip between two probability game modes and so have the potential to both maintain and heal skin, challenging the view that only rare stem cells matter. [Press release from the Wellcome Trust Sanger Institute (EurekAlert!) discussing online prepublication in Nature Cell Biology] Press Release | Abstract

DERMAL STEM CELLS & TISSUE REGENERATION

Bone Marrow Mesenchymal Stem Cell Aggregate: An Optimal Cell Therapy for Full-Layer Cutaneous Wound Vascularization and Regeneration
Cell-aggregates constructed with green fluorescent protein-expressing rat bone marrow mesenchymal stem cells were applied to rat acute full-layer cutaneous wound model to confirm its pro-regeneration ability and compare its regenerative efficacy with the currently thriving subcutaneous and intravenous stem cell administration strategy, with a view to sensing the advantages, disadvantages and the mechanism behind. Hyaluronan Does Not Regulate Human Epidermal Keratinocyte Proliferation and Differentiation
To investigate hyaluronan (HA) functions, scientists studied the effects of HA depletion on human keratinocyte physiology within in vitro reconstructed human epidermis. [J Biol Chem] Abstract | Full Article

Cellular Events during Scar-Free Skin Regeneration in the Spiny Mouse, Acomys
Investigators compared the responses of scarring and non-scarring organisms concentrating on the immune cells and wound cytokines, cell proliferation and the collagenous components of the wound bed and scar. [Wound Repair Regen] Abstract

Melanocyte Antigen Triggers Autoimmunity in Human Psoriasis
Researchers identified melanocytes as skin-specific target cells of an human leukocyte antigen (HLA)-C*06:02–restricted psoriatic T cell response. They found that a Vα3S1/Vβ13S1 T cell receptor specifically recognized HLA-C*06:02–positive melanocytes. [J Exp Med] Abstract

Donor-Derived Stem-Cells and Epithelial Mesenchymal Transition in Squamous Cell Carcinoma in Transplant Recipients
Investigators studied stem-cells and epithelial mesenchymal transition expression in skin squamous-cell-carcinomas and concomitant actinic keratoses in kidney-transplant recipients. [Oncotarget] Full Article

A Synthetic Modular Approach for Modeling the Role of the 3D Microenvironment in Tumor Progression
Tumor cell function was investigated by culturing WM239A melanoma cells on poly(ethylene glycol) hydrogel surfaces or encapsulating cells within the hydrogels, and either as monocultures or indirect cocultures with primary human dermal fibroblasts. [Sci Rep] Full Article

Downregulation of Pyrroline-5-Carboxylate Reductase-2 Induces the Autophagy of Melanoma Cells via AMPK/mTOR Pathway
Investigators focused on the effect of pyrroline-5-carboxylate reductase-2 (PYCR2) on inducing autophagy process in melanoma. They found that silencing PYCR2 resulted in the decrease of proliferative ability and activation of AMPK/mTOR-induced autophagy of A375 cells. [Tumor Biol] Abstract

Master Regulators of Infiltrate Recruitment in Autoimmune Disease Identified through Network-Based Molecular Deconvolution
Using alopecia areata as a model, investigators adapted regulatory network analysis to specifically isolate physiological behaviors in the skin that contribute to the recruitment of immune cells in autoimmune disease. [Cell Syst] Full Article | Graphical Abstract

Collagen Interactions: Drug Design and Delivery
Strategies to target collagen interactions are already being developed, including the use of monoclonal antibodies to interfere with collagen fibril formation and the use of triple-helical peptides to direct liposomes to melanoma cells. Intelligent control of collagen biological interactions within a material context will expand the effectiveness of collagen-based drug delivery. [Adv Drug Deliv Rev] Abstract
