TOP STORYT Follicular Helper-Like Cells Contribute to Skin Fibrosis Researchers showed that CD4+ inducible costimulator (ICOS)-expressing T cells with a T follicular helper (TFH) -like phenotype infiltrate the skin of patients with systemic sclerosis (SSc) and are correlated with dermal fibrosis and clinical disease status. ICOS+ TFH-like cells were found to be increased in the skin of graft-versus-host disease–SSc mice and contributed to dermal fibrosis via an interleukin-21- and matrix metalloproteinase 12–dependent mechanism. PUBLICATIONS(Ranked by impact factor of the journal)DERMAL STEM CELLS & TISSUE REGENERATIONp63 Is a Key Regulator of iRHOM2 Signaling in the Keratinocyte Stress Response The authors report that iRHOM2 is a novel target gene of p63 and that both p63 and iRHOM2 differentially regulate cellular stress-associated signaling pathways in normal and hyperproliferative keratinocytes. They demonstrated that p63–iRHOM2 regulates cell survival and response to oxidative stress via modulation of SURVIVIN and Cytoglobin, respectively. [Nat Commun] Full Article Investigators elucidated the underlying mechanism of non-thermal atmospheric pressure plasma (NTAPP)-induced interleukin-8 (IL-8) expression in human keratinocyte HaCaT cells. [Arch Biochem Biophys] Abstract Macrophage-Derived GPNMB Accelerates Skin Healing Transplantation of macrophages expressing GPNMB improves skin healing in GPNMB-mutant mice. Additionally, topical treatment with recombinant GPNMB restored mesenchymal stem cells recruitment and accelerated wound closure in the diabetic skin. [Exp Dermatol] Abstract | Full Article SKIN CANCERS & DISORDERSInvestigators found that metformin attenuates melanoma growth and metastasis by reducing TRIB3 expression in non-diabetic C57BL/6 mice and diabetic KK-Ay mice; overexpression of TRIB3 protects metformin from the activation of autophagic flux, the clearance of accumulated tumor-promoting factors and the attenuation of tumor progression. [Oncogene] Abstract Uncovering and Deciphering the Pro-Invasive Role of HACE1 in Melanoma Cells Scientists demonstrated that HACE1 promoted melanoma cell migration and adhesion in vitro and was required for mouse lung colonization by melanoma cells in vivo. [Cell Death Differ] Abstract Loss of miR-204 Expression Is a Key Event in Melanoma MiR-204 represents a relevant mechanism in melanoma, with potential prognostic value and its loss seems to act in the CDKN2A pathway, in cooperation with NRAS. [Mol Cancer] Full Article Down-Regulation of GADD45A Enhances Chemosensitivity in Melanoma Researchers investigated the role of GADD45A in chemotherapy response. They found the expression of GADD45A upregulated in a dose- and time- dependent manner. Suppression of GADD45A sensitized melanoma cells to cisplatin and enhanced cisplatin-induced DNA damage. [Sci Rep] Full Article Scientists evaluated the possible inhibitory effect of Notch1 signaling inhibitor, γ-secretase inhibitor DAPT, on psoriatic Th17 cell differentiation and function in a mouse model of psoriasis-like skin inflammation. [J Transl Med] Full Article Investigators analyzed, in keratinocytes, the transcriptomic effect of FUCA1 knock-down for a better understanding of skin lesions’ pathogenesis affecting fucosidosis patients. REVIEWSInterleukin-31 and Interleukin-31 Receptor – New Therapeutic Targets for Atopic Dermatitis IL-31 receptor α-chain (IL-31RA) is expressed in peripheral nerves and epidermal keratinocytes, and the role of IL-31 on the pruritus and skin barrier have been investigated. An anti-IL-31 receptor antibody was shown to significantly improve pruritus in atopic dermatitis patients. POLICY NEWSKey Republican Lawmaker Releases New ‘Right-to-Try’ Legislation A key Republican lawmaker released a new version of so-called “right to try” legislation, a pivotal next step in the effort to help dying patients get access to experimental therapies. [STAT News] Editorial Scientists Win and Lose in Texas Primary Contests Texas primary was the first test for scientists seeking seats in the U.S. House of Representatives, and the results were mixed. [ScienceInsider] Editorial Behavioral ‘Violations’ Cost Prominent Neuroscientist Positions at Columbia, HHMI Prominent Columbia University neuroscientist Tom Jessell, 66, has been fired for “serious [behavioral] violations” and the university is closing his lab, the New York City institution said in a statement. The Howard Hughes Medical Institute (HHMI) in Chevy Chase, Maryland, which effectively employed Jessell, a leader in understanding neural development, also terminated him as an investigator because he “violated HHMI policy,” according to a statement from the institute. [ScienceInsider] Editorial Police Probe of Brazilian Marijuana Researcher Sparks Protests A police investigation targeting Brazil’s most prominent marijuana researcher has ignited a wave of protest among scientists. They say that the move by authorities from the state of São Paulo threatens research freedoms at a time when science in the country faces severe problems because of draconian budget cuts.
