TOP STORYSenescent Human Melanocytes Drive Skin Aging via Paracrine Telomere Dysfunction Scientists showed that melanocytes were the only epidermal cell type to express the senescence marker p16INK4A during human skin aging. Aged melanocytes also displayed additional markers of senescence such as reduced HMGB1 and dysfunctional telomeres, without detectable telomere shortening. [EMBO J] Abstract

PUBLICATIONS(Ranked by impact factor of the journal)DERMAL STEM CELLS & TISSUE REGENERATIONInvestigators showed that autologous micrograft (AMG) improved skin re-epithelialization by accelerating the migration of fibroblasts and keratinocytes. More specifically, AMG-treated wounds showed improvement of indispensable events associated with successful wound healing such as granulation tissue formation, organized collagen content, and newly formed blood vessels. [Cell Death Differ] Abstract Millimeter-Wave Pulsed Heating In Vitro: Cell Mortality and Heat Shock Response Scientists evaluated apoptotic and heat shock responses of melanoma cells exposed in vitro to continuous or pulsed-wave amplitude-modulated MMW at 58.4 GHz with the same average temperature rise. Using an ad hoc exposure system, they generated 90 minute pulse trains with 1.5 second pulse duration, period of 20 seconds, amplitude of 10°C, and steady-state temperature at the level of cells of 49.2°C. [Sci Rep] Full Article In nitrogen-mustard-exposed mouse skin, scientists found p-ERK activation increased Fra-1 and FosB. Unlike the nucleus location of c-Fos and FosB, Fra-1 and Fra-2 were mainly expressed in the cytoplasm. In nitrogen-mustard-exposed cultured immortalized human keratinocytes, Fra-1 in the nucleus functioned as an inhibitor of inflammatory cytokine interleukin IL-8. [Toxicol Lett] Abstract Effective silk fibroin dosage was determined and incorporated according to minimum inhibitory concentrations against wound associated bacterial strains while fabricating scaffold. Dermal fibroblast NIH3T3 cells were seeded on epidermal growth factor treated and untreated polyurethane/silk fibroin scaffolds, fabricated by blending and immobilization techniques. [Int J Biol Macromol] Abstract | Graphical Abstract Investigators evaluated human dermal papilla cells viability, alkaline phosphatase activity, protein expression, and vascular endothelial growth factor secretion in vitro and assessed the anagen-promoting effects of boehmite via gross observation and histological analysis in a mouse model. [Exp Dermatol] Abstract SKIN CANCERS & DISORDERSThrough pharmacological and genetic approaches, the authors showed that GABA-A acts through calcium signaling to maintain melanocyte stem cell quiescence. Unexpectedly, the authors identified translocator protein (TSPO), a mitochondrial membrane associated protein that regulated mitochondrial function and metabolic homeostasis, as a parallel regulator of melanocyte stem cell quiescence. [Pigment Cell Melanoma Res] Abstract Expression Level of EGFR and MET Receptors Regulates Invasiveness of Melanoma Cells Researchers analyzed direct involvement of these receptors in the invasion of melanoma cells inducing EGFR and MET up- and down-regulations in examined cells. Results were acquired with assays evaluating cell migration and invasion (scratch wound assay, Transwell filter-based method and single-cell tracking). [J Cell Mol Med] Full Article Small Extracellular Vesicles Convey the Stress-Induced Adaptive Responses of Melanoma Cells Investigators compared B16F1 cell-derived small extracellular vesicles (sEVs) released under normal and stress conditions, including cytostatic, heat and oxidative stress. The miRNome and proteome showed substantial differences across sEV groups and bioinformatics analysis of the obtained data by the Ingenuity Pathway Analysis also revealed significant functional differences. [Sci Rep] Full Article Researchers first showed that the expression levels of RNF6 in multiple myeloma were significantly elevated compared with the bone marrow cells of healthy donors. Overexpression of RNF6 in LP1 and PRMI-8266 multiple myeloma cell lines promoted cell proliferation, whereas knockdown of RNF6 led to apoptosis of multiple myeloma cells. REVIEWSNew and Evolving Concepts of Melanocytic Nevi and Melanocytomas Recently, many of the mutations that initiate nevus formation have been identified and specific sets of mutations are found in different subtypes of nevi. The author provides an update on the broad spectrum of melanocytic nevi and melanocytomas and outlines their key histopathologic and genetic features. [Mod Pathol] Abstract Immune-Competent In Vitro Co-culture Models as an Approach for Skin Sensitization Assessment Significant progress has been made over the past decades to develop new technologies of data generation concurrently with novel approaches to improve the models especially by the use of co-culture. Scientists address the technical aspects to take into consideration when co-culturing depending on the desired objective and describes the different keratinocytes and dendritic cells co-cultures developed in 2D and 3D. INDUSTRY NEWSMOR106 Clinical Development in Atopic Dermatitis Stopped for Futility MorphoSys AG and Galapagos NV announced the end of the clinical development program of MOR106 in atopic dermatitis. The joint decision of all three involved parties, Galapagos NV, MorphoSys AG and Novartis Pharma AG, was based on an interim analysis for futility that was performed in the Phase II IGUANA trial. [MorphoSys AG] Press Release DermTech Receives CPT® Proprietary Laboratory Assay Code from the American Medical Association DermTech, Inc. announced the American Medical Association has issued a Proprietary Laboratory Analyses (PLA) Common Procedural Terminology (CPT®) Code for the DermTech Pigmented Lesion Assay. This PLA code was published online with an effective date of July 1, 2019 and is included in the CPT 2020 publication. [DermTech, Inc. (GlobeNewswire, Inc.)] Press Release

POLICY NEWSUCL to Phase Out Single-Use Plastics, Including Pipette Tips University College London made a series of pledges last week to cut down on plastic waste, help mitigate climate change, and make its research and teaching facilities more environmentally sustainable, according to a press release issued on October 16. If all goes to plan, UCL will divest from fossil fuel-producing companies by the end of this year, have one and a half football pitches’ worth of green space on its campus within five years, and offer vegetarian options only through its catering and hospitality services by 2030.
