TOP STORYHair-Bearing Human Skin Generated Entirely from Pluripotent Stem Cells Scientists report an organoid culture system that generated complex skin from human pluripotent stem cells. They used stepwise modulation of the transforming growth factor β and fibroblast growth factor signaling pathways to co-induce cranial epithelial cells and neural crest cells within a spherical cell aggregate. PUBLICATIONS(Ranked by impact factor of the journal)DERMAL STEM CELLS & TISSUE REGENERATIONResearchers applied an experimental platform for melanoma cell lines that leveraged recombinant heavy isotope-coded peptide MHCs and multiplex isotope tagging. They profiled the immunopeptidome response to CDK4/6 inhibition and interferon-γ. [Nat Commun] Full Article Investigators designed a gene therapy strategy using the adenovirus-mediated human laminin-332 α3 chain (LAMA3) gene transduced into a human immortalized epidermal cell line to study the formation of the hemidesmosome in vitro. [Mol Ther Methods Clin Dev] Full Article | Graphical Abstract Deferiprone Stimulates Aged Dermal Fibroblasts via HIF-1α Modulation The authors analyzed the efficacy of the iron-chelator Deferiprone (DFP) to stimulate dermal fibroblasts in-vitro and correlated the effective DFP concentrations with its ability to penetrate the epidermis, reach the dermis and activate HIF-1α in-vivo. [Aesthet Surg J] Abstract SKIN CANCERS & DISORDERSScientists showed that when melanoma cells were exposed to the aged fibroblast lipid secretome, or co-cultured with aged fibroblasts, they increased the uptake of lipids, via the fatty acid transporter, fatty acid transport protein (FATP) 2. [Cancer Discov] Abstract The Tumor Microenvironment Shapes Dendritic Cell Plasticity in a Human Organotypic Melanoma Culture Researchers developed a human organotypic skin melanoma culture that allowed real-time study of host-malignant cell interactions within a multicellular tissue architecture. This provided a valuable approach to study how a tumor microenvironment can affect the immune system. [Nat Commun] Full Article Investigators report that BAX-interacting protein 1 (BIF-1) was significantly downregulated in both primary and metastatic melanomas, and that patients with high levels of BIF-1 expression exhibited a better overall survival. Depleting BIF-1 resulted in higher proliferation rates both in vitro and in vivo. [Oncogene] Abstract Antitumor Effect of Cell Therapy with Mesenchymal Stem Cells on Murine Melanoma B16-F10 Researchers investigated the antitumor and immunomodulatory effects of mesenchymal stem cells (MSC) obtained from bone marrow in the treatment of dorsal melanoma B16-F10. They suggested that undifferentiated MSC, administered intratumor and intravenous is possibly a promising treatment for melanoma. [Biomed Pharmacother] Full Article | Graphical Abstract The authors found that inhibition of the MAPK pathway activated the JAK2/STAT3 pathway, while JAK2 inhibitors that inhibit the JAK2/STAT3 pathway activated the MAPK pathway, suggesting crosstalk between these two pathways in BRAF mutant melanoma cells. [Mol Ther Oncolytics] Full Article | Graphical Abstract Scientists elucidated the role of the histone methyltransferase SETDB1 in melanoma, showing that SETDB1 modulated relevant transcriptomic effects in melanoma, in particular as activator of cancer-related secreted factors and as repressor of melanocyte-lineage differentiation and metabolic enzymes. POLICY NEWSNIH Grapples with Rush to Claim Billions in Pandemic Research Funds For the second time in just over ten years, the National Institutes of Health (NIH) is scrambling to hand out billions of dollars in emergency research funding and scientists are rushing to get a piece of the action, even as some confusion and concerns abound. [ScienceInsider] Editorial As the Pandemic Hits Campus Finances, Faculty Face Layoffs The pandemic has bored holes in budgets at universities and colleges across the US and beyond. To stem the spread of the coronavirus, higher education institutions shuttered their campuses, sent students home, and moved classes online. [The Scientist] Editorial
