Galectin-7 Downregulation in Lesional Keratinocytes Contributes to Enhanced IL-17A Signaling and Skin Pathology in Psoriasis

Scientists found that the expression of galectin-7 was reduced in the skin lesions of patients with psoriasis. IL-17A and TNF-α, two cytokines intimately involved in the development of psoriatic lesions, suppressed galectin-7 expression in human primary keratinocytes and the immortalized human keratinocyte cell line HaCaT.

Contribution of GATA6 to Homeostasis of the Human Upper Pilosebaceous Unit and Acne Pathogenesis

GATA6 was down-regulated in acne when expressed in the upper pilosebaceous unit of normal human skin. GATA6 controls keratinocyte proliferation and differentiation to prevent hyperkeratinization of the infundibulum, which is the primary pathological event in acne.

Dissecting the Roles of GRK2 and GRK3 in μ-Opioid Receptor Internalization and β-Arrestin2 Recruitment Using CRISPR/Cas9-Edited HEK293 Cells

Using CRISPR/Cas9-mediated genome editing, researchers established HEK293 cells with knockout of GRK2, GRK3 or both to dissect their individual contributions in β-arrestin2 recruitment and μ-OR internalization upon stimulation with four different agonists.

The Long Noncoding RNA PDK1-AS/miR-125b-5p/VEGFA Axis Modulates Human Dermal Microvascular Endothelial Cell and Human Umbilical Vein Endothelial Cell Angiogenesis after Thermal Injury

Investigators identified the role of long noncoding RNAs related to the function of miR‐125b and vascular endothelial growth factor in burn wound repair and the underlying mechanism.

Coordination Between Cell Motility and Cell Cycle Progression in Keratinocyte Sheets via Cell-Cell Adhesion and Rac1

Scientists found that keratinocyte sheets exhibited time-dependent coordination of collective cell movement and cell cycle progression after seeding cells.

WNT11-FZD7-DAAM1 Signaling Supports Tumor Initiating Abilities and Melanoma Amoeboid Invasion

Researchers showed that WNT11-FZD7-DAAM1 activated Rho-ROCK1/2-Myosin II and played a crucial role in regulating tumor-initiating potential, local invasion and distant metastasis formation.

Enhancer Reprogramming Confers Dependence on Glycolysis and IGF Signaling in KMT2D Mutant Melanoma

The authors identified KMT2D as a potent tumor suppressor in melanoma through an in vivo epigenome-focused pooled RNAi screen and confirmed the finding by using a genetically engineered mouse model based on conditional and melanocyte-specific deletion of KMT2D.

Melanoma Stem Cell-Like Phenotype and Significant Suppression of Immune Response within a Tumor Are Regulated by TRIM28 Protein

TRIM28 high level related to higher melanoma stemness as measured with several distinct scores and TRIM28HIGH-expressing melanoma cell lines possess the greater potential of melanosphere formation.

Inhibition of Melanoma Cell Migration and Invasion Targeting the Hypoxic Tumor Associated CAXII

Researchers demonstrated that malignant melanoma cell motility was impaired by targeting carbonic anhydrase IX via either carbonic anhydrase inhibitors or through the inhibition of the Hedgehog pathway.

Response of Human Melanoma Cell Lines to Interferon-Beta Gene Transfer Mediated by a Modified Adenoviral Vector

The AdRGD-PG-hIFNβ vector encoding the human interferon-β cDNA was used to transduce human melanoma cell lines SK-MEL-05 and SK-MEL-147. In vitro, cell death was induced in more than 80% of the cells and correlated with elevated annexinV staining and caspase 3/7 activity.
