Network Models of Primary Melanoma Microenvironments Identify Key Melanoma Regulators Underlying Prognosis

Seventeen key drivers of the gene subnetworks associated with poor prognosis, including the transcription factor ZNF180, were tested for their pro-tumorigenic effects in vitro. The anti-tumor effect of silencing ZNF180 was further validated using in vivo xenografts.

Identification and Characterization of IMD-0354 as a Glutamine Carrier Protein Inhibitor in Melanoma

IMD-0354 was confirmed as a potent inhibitor of glutamine uptake that attained sustained low intracellular glutamine levels. Concomitant with its inhibition of glutamine uptake, IMD-0354 attenuated mTOR signaling, suppressed 2D and 3D growth of melanoma cells, and induced cell cycle arrest, autophagy and apoptosis.

Transcriptional Network Constituted of CBP, Ku70, NOX2, and BAX Prevents the Cell Death of Necrosis, Paraptosis, and Apoptosis in Human Melanoma

Knocking down CREB-binding protein or Ku70 induced necrotic and paraptotic cell death as indicated by high-level intracellular reactive oxygen species, cytoplasmic vacuolization, and cell cycle arrest in the S phase.

Inhibition of Phospholipases Suppresses Progression of Psoriasis through Modulation of Inflammation

Investigators transfected si-PLA2G4B in a murine keratinocyte cell-line PAM212 to verify the effect of progression by PLA2G4B.

Screening of Metabolic Modulators Identifies New Strategies to Target Metabolic Reprogramming in Melanoma

To support the discovery of new drug candidates for melanoma, scientists examined 180 metabolic modulators, including phytochemicals and anti-diabetic compounds, for their growth-inhibitory activities against melanoma cells, alone and in combination with the BRAF inhibitor vemurafenib. 4-methylumbelliferone and ursolic acid, were subjected to validation and further characterization.

Mechanical Stretch Promotes Hypertrophic Scar Formation through Mechanically Activated Cation Channel Piezo1

In vitro, cyclic mechanical stretch (CMS) increased Piezo1 expression and Piezo1-mediated calcium influx in human dermal fibroblasts (HDFs). Piezo1 activity promoted HDF proliferation, motility, and differentiation in response to CMS. Intradermal injection of GsMTx4, a Piezo1-blocking peptide, protected rats from stretch-induced hypertrophic scar formation.

A Wnt5a-Cdc42 Axis Controls Aging and Rejuvenation of Hair-Follicle Stem Cells

Scientists showed that aged hair follicle stem cells present with a decrease in canonical Wnt signaling and a shift towards non-canonical Wnt5a drove signaling which antagonized canonical Wnt signaling.

A Surfactant Polymer Wound Dressing Protects Human Keratinocytes from Inducible Necroptosis

Researchers showed that TX100-induced plasma membrane damage and death by necroptosis were both attenuated by surfactant polymer dressing, allowing keratinocyte survival.

Structural Characteristics of Polysaccharide Microcapsules from Nostoc commune, and Their Applications in Skin Wound Healing and Pathological Repair

The extraction conditions of Nostoc commune Vauch polysaccharide (NCVP) were optimized by single factor and orthogonal experiments. Then, microcapsules (NCVPM) were prepared. After analyzing the microcapsule structural and thermal characteristics, the skin wound healing ability was studied by establishing back trauma rat models.
