TOP STORYJQ1 prevented hypoxia-inducible factor (HIF) binding to the hypoxia response element in carbonic anhydrase 9 (CA9) promoter, but did not alter HIF expression or activity, suggesting some HIF targets are bromodomain and extra-terminal (BET)-dependent. JQ1 reduced triple negative breast cancer growth in vitro and in vivo and inhibited xenograft vascularization. [Oncogene] Full Article | Press Release

PUBLICATIONS(Ranked by impact factor of the journal)Scientists showed that tumor endothelial cells (TECs) actively promote tumor metastasis through a bidirectional interaction between tumor cells and TECs. Co-implantation of TECs isolated from highly metastatic tumors accelerated lung metastases of low metastatic tumors. [Sci Rep] Full Article Estrogen Stimulates Homing of Endothelial Progenitor Cells to Endometriotic Lesions The incorporation of endothelial progenitor cells (EPCs) into microvessels contributes to the vascularization of endometriotic lesions. Investigators analyzed whether this vasculogenic process is regulated by estrogen. Estrogen- and vehicle-treated human EPCs were analyzed for migration and tube formation. [Am J Pathol] Abstract Scientists tested the hypothesis that metformin could accelerate wound healing by improving the impaired endothelial progenitor cell functions in streptozotocin-induced diabetic mice. [Cardiovasc Diabetol] Full Article Scientists studied whether the histone deacetylase inhibitor trichostatin A affects NADPH oxidase 4 (Nox4) expression and angiogenesis. Nox4 expression was measured by real time PCR and Western blot analysis in endothelial cells. [J Cell Mol Med] Full Article The Role of Twist1 Phosphorylation in Angiogenesis and Pulmonary Fibrosis The authors showed that a transcription factor, Twist1, controls angiogenesis through the angiopoietin-Tie2 pathway and deregulation of this mechanism mediates pathological angiogenesis and collagen deposition in a bleomycin-induced mouse pulmonary fibrosis model. [Am J Respir Cell Mol Biol] Abstract The authors determined if exposure to fine and ultrafine particles from indoor and outdoor sources, assessed by personal and residential indoor monitoring, is associated with altered levels of endothelial progenitor cells, and whether such effects are related to leukocyte-mediated oxidative stress. [Toxicology] Full Article Regulation of Retinal Angiogenesis by Phospholipase C-β3 Signaling Pathway Researchers showed that phospholipase C-β3 (PLC-β3) isoform regulates endothelial cell function and retinal angiogenesis. Silencing of PLC-β3 in human umbilical vein endothelial cells significantly delayed proliferation, migration and capillary-like tube formation. [Exp Mol Med] Full Article Researchers focused on the effects of acidosis on lymphatic endothelial cells (LECs) to assess the relationship between acidic microenvironments and lymph node metastasis. They demonstrated that normal human LECs express various acid receptors by immunohistochemistry and reverse transcriptase-polymerase chain reaction. [Exp Cell Res] Abstract Scientists investigated the anti-angiogenic potential of daphnetin (DAP), focusing on the mechanism of action. In vivo anti-angiogenic potential of DAP was evaluated by vascular endothelial growth factor (VEGF)-induced rat aortic ring assay and chick chorioallantoic membrane assay. [Clin Exp Pharmacol Physiol] Abstract Investigators showed for the first time that both human dermal microvascular endothelial cells (HDMECs) and human brain microvascular endothelial cell (HBMECs) express α-adrenergic receptors. Moreover, they indicated that phentolamine inhibits the proliferation, migration, and tubulogenesis of HDMECs and HBMECs. [Clin Hemorheol Microcirc] Abstract

REVIEWSRole of PGC-1α in Vascular Regulation: Implications for Atherosclerosis The authors detail recent findings related to peroxisome proliferator receptor-γ coactivator 1α (PGC-1α) in vascular regulation, regulation of PGC-1α itself, the role of PGC-1α in atherosclerosis, and therapies that target this key protein. [Arterioscler Thromb Vasc Biol] Abstract Vascular CaMKII: Heart and Brain in Your Arteries The authors focus on emerging evidence of multifunctional calcium/calmodulin-dependent protein kinase II (CaMKII) as an essential component of the vascular system, with a focus on the kinase isoform/splice variants and cellular system studied. INDUSTRY NEWSOphthotech Corporation announced the completion of patient recruitment in its Phase III trial of Fovista®, anti-PDGF therapy, in combination with Eylea® or Avastin® for the treatment of wet age-related macular degeneration. [Ophthotech Corporation] Press Release Genomic Health Launches Oncotype SEQ™ Liquid Biopsy Mutation Panel Genomic Health, Inc. announced the commercial launch of Oncotype SEQ™ Liquid Select, the first of several non-invasive liquid biopsy tests that the company plans to deliver through its Oncotype IQ™ Genomic Intelligence Platform. Oncotype SEQ is a blood-based test that uses next-generation sequencing to identify and assess actionable genomic alterations in a panel of 17 select genes to inform the treatment of stage IV solid tumors, including lung, breast, colon, melanoma, ovarian, and gastrointestinal stromal tumors.
