    Local Mitochondrial ATP Production Regulates Endothelial Fatty Acid Uptake and Transport

    Inhibitors of oxidative phosphorylation and the ATP/ADP translocase suppressed FA uptake, pointing principally to ATP production. Decreasing total cellular ATP by blocking glycolysis did not decrease uptake, indicating that specifically mitochondrial ATP was required.

    Blockade of VEGF-C Signaling Inhibits Lymphatic Malformations Driven by Oncogenic PIK3CA Mutation

    Investigators showed that the somatic PIK3CAH1047R mutation, resulting in constitutive activation of the p110α PI3K, underlaid both macrocystic and microcystic lymphatic malformations (LMs) in human. Using a mouse model of PIK3CAH1047R-driven LMs, they demonstrated that both types of malformations arose due to lymphatic endothelial cell-autonomous defects, with the developmental timing of p110α activation determining the LM subtype.

    Angiopoietin-2–Integrin α5β1 Signaling Enhances Vascular Fatty Acid Transport and Prevents Ectopic Lipid-Induced Insulin Resistance

    Mechanistically, Angpt2 activated integrin α5β1 signaling in the endothelium and triggered fatty acid transport via CD36 and FATP3 into subcutaneous adipose tissue. Genetic or pharmacological inhibition of the endothelial integrin α5β1 recapitulated adipocyte-specific Angpt2 knockout phenotypes.

    Myeloid ALX/FPR2 Regulates Vascularization Following Tissue Injury

    Mechanistically, RNA sequencing revealed that resolvin D1 induced a transcriptional program in macrophages characteristic of a pro-revascularization phenotype. Vascularization of ischemic skeletal muscle, as well as cutaneous wounds, was impaired in mice with myeloid-specific deficiency of Alx/Fpr2, and this was associated with altered expression of pro-revascularization genes in skeletal muscle and macrophages isolated from skeletal muscle.

    Pro-Lymphangiogenic VEGFR-3 Signaling Modulates Memory T Cell Responses in Allergic Airway Inflammation

    Researchers explored how pro-lymphangiogenic signaling affects the allergic response to house dust mite. In the acute inflammatory phase, the lungs of mice treated with blocking antibodies against VEGFR-3 displayed less inflammation overall, with dramatically reduced innate and T-cell numbers and reduced inflammatory chemokine levels.

    Cell Type Specific Adhesion to Surfaces Functionalised by Amine Plasma Polymers

    The increased resistance observed for the non-endothelial cell types was accompanied by an increased rate of cellular attachment, even though spontaneous migration was comparable to the control, i.e., to the standard cultivation surface. As demonstrated on fibroblasts, the resistance to trypsin was similar in serum-supplemented and serum-free media.

    miR-503-5p Inhibits Colon Cancer Tumorigenesis, Angiogenesis, and Lymphangiogenesis by Directly Downregulating VEGF-A

    Scientists observed considerable downregulation of miR-503-5p expression in colon cancer cells and tissues and significant correlation with the TNM stage, differentiation grade and lymph node metastasis of colon cancer. Overexpression of miR-503-5p promoted the apoptosis and G1 arrest of colon cancer cells, and inhibited migration, proliferation, invasion and colony formation.
