Signaling Promotes the Development of Endocardial Cells from Human Pluripotent Stem Cell-Derived Cardiovascular Progenitors

To access human endocardial cells, investigators generated a human PSC-derived endothelial population that displayed many characteristics of endocardium, including expression of the cohort of genes that identifies this lineage in vivo, the capacity to induce a trabecular fate in immature cardiomyocytes in vitro, and the ability to undergo EMT.

p300 Promotes Portal Hypertension and Hepatic Fibrosis through CCL2-Mediated Angiocrine Signaling

In vitro, epigenome‐editing of C‐C motif chemokine ligand 2 (CCL2) enhancer and promoter regions by CRISPR‐dCas9‐KRAB technology repressed TNFα‐induced CCL2 transcription via H3K9 tri‐methylation.

    Mapping
Endothelial-Cell Diversity in Cerebral Cavernous Malformations at Single-Cell Resolution

Investigators report in-depth single-cell RNA sequencing, combined with spatial transcriptomics and immunohistochemistry, to comprehensively characterize subclasses of brain endothelial cells under both normal conditions and after deletion of Pdcd10 in a mouse model of cerebral cavernous malformation.

Marrow Stromal Cells Stimulated by Strontium-Substituted Calcium Silicate Ceramics: Release of Exosomal miR-146a Regulates Osteogenesis and Angiogenesis

Researchers showed that bone marrow stem cell-derived exosomes after strontium-substituted calcium silicate stimulation could significantly promote in vitro angiogenesis of human umbilical vein endothelial cells, which might be attributed to elevated pro-angiogenic miR-146a cargos and inhibition of Smad4 and NF2 proteins.

Carried by Human Umbilical Cord Mesenchymal Stem Cell Enhances Endothelial Function through Exosome-Mediated Mechanisms In Vitro and Attenuates Vein Graft Neointimal Formation In Vivo

Human umbilical cord MSCs (hucMSCs) and HUVECs were isolated from human umbilical cords and characterized by a series of experiments. Lentivirus vector encoding miRNA-126-3p was transfected into hucMSCs and verified by PCR.

    ZNF354C
Is a Transcriptional Repressor That Inhibits Endothelial Angiogenic Sprouting

Overexpression of ZNF354C in human endothelial cells results in a marked inhibition of endothelial sprouting. RNA-sequencing of human microvascular endothelial cells with and without overexpression of ZNF354C revealed that the protein is a potent transcriptional repressor.

    Expression
of CARD8 in Human Atherosclerosis and Its Regulation of Inflammatory Proteins in Human Endothelial Cells

The CARD8 mRNA was knocked-down in HUVECs in vitro, followed by quantitative RT-PCR analysis and OLINK Proteomics. Endothelial and smooth muscle cells in arterial tissue expressed CARD8 and CARD8 correlated with vWF, CD163 and the expression of inflammatory genes, such as CXCL1, CXCL6 and PDGF-A in plaque.

Kinase M2 Mediates the Glycolysis in the Lymphatic Endothelial Cells and Its Potential Relation to the Progression of Lymphatic Malformations

The glycolysis index, including glucose uptake, ATP and lactate production stayed at a relatively high level in human dermal lymphatic endothelial cells (HDLECs) compared with HUVECS, while inhibition of PKM2 by shikinon or PKM2 knockdown could significantly suppress glycolysis, migration, tubular formation, and invasion of HDLECs.

Acid Reverses the Promoting Effects of Breast Tumor Cell-Derived Exosomes on Endothelial Cell Migration and Angiogenesis

While treatment with exosomes obtained from untreated breast cancer cells, tumor cell-derived exosomes (TDE), enhanced the expression of vVEGF-A in human umbilical vein endothelial cells, incubation with DHA or TDE led to the significant decrease of VEGF-A transcript level in these cells.
