Highly Efficient Cas9-Mediated Transcriptional Programming
The authors describe an improved transcriptional regulator obtained through the rational design of a tripartite activator, VP64-p65-Rta, fused to nuclease-null Cas9. They demonstrate its utility in activating endogenous coding and noncoding genes, targeting several genes simultaneously and stimulating neuronal differentiation of human induced pluripotent stem cells. Protective Effects of Human iPS-Derived Retinal Pigmented Epithelial Cells in Comparison with Human Mesenchymal Stromal Cells and Human Neural Stem Cells on the Degenerating Retina in rd1 Mice
Researchers investigated the rescue effects of induced pluripotent stem-retinal pigmented epithelium cells in comparison with human mesenchymal stromal cells and human neural stem cells in a clinical trial on photoreceptor degeneration in rd1 mice. [Stem Cells] Abstract

p53 Is Active in Murine Stem Cells and Alters the Transcriptome in a Manner that Is Reminiscent of Mutant P53
The authors showed that a significant part of p53 is localized in the nucleus of murine embryonic stem cells and that the majority of this nuclear p53 is bound to DNA. According to its nuclear localization, they showed that p53 alters the transcriptional program of stem cells. [Cell Death Dis] Full Article

Induced Pluripotent Stem Cells as a Model for Diabetes Investigation
Researchers aimed to generate and evaluate differentiation potential of induced pluripotent stem cells from lepdb/db mice, the model of diabetes type 2 as well as from patients with maturity onset diabetes of the young 3. [Sci Rep] Full Article

GSK3 Inhibitors CHIR99021 and 6-Bromoindirubin-3′-Oxime Inhibit MicroRNA Maturation in Mouse Embryonic Stem Cells
To investigate the effect of GSK3 inhibitors on microRNA (miRNA) expression, investigators analyzed the miRNA expression profile of J1 mouse embryonic stem cells in the absence or presence of CHIR99021 or 6-bromoindirubin-3′-oxime by small RNA deep-sequencing. [Sci Rep] Full Article

Increased Risk of Genetic and Epigenetic Instability in Human Embryonic Stem Cells Associated with Specific Culture Conditions
Scientists performed a combinatorial study on both human embryonic stem cells and human induced pluripotent stem cells to compare the effects of enzymatic vs. mechanical passaging, and feeder-free vs. mouse embryonic fibroblast feeder substrate, on the genetic and epigenetic stability and the phenotypic characteristics of human pluripotent stem cells. Preferential Gene Expression and Epigenetic Memory of Induced Pluripotent Stem Cells Derived from Mouse Pancreas
Investigators established induced pluripotent stem cells (iPSCs) derived from mouse pancreas (Panc-iPSC) and compared their properties with those of iPSC derived from tail-tip fibroblast (TTF-iPSC). The metabolic profile differs between Panc-iPSC and TTF-iPSC, indicating distinct cell properties in these iPSCs. [Genes Cells] Abstract

Oct3/4 Directly Regulates Expression of E2F3a in Mouse Embryonic Stem Cells
Scientists identified E2F3a as a direct target gene of Oct3/4 in embryonic stem (ES) cells. Oct3/4 directly bound to the promoter region of the E2F3a gene and positively regulated expression of E2F3a in mouse ES cells. [Biochem Biophys Res Commun] Abstract

The L1TD1 Protein Interactome Reveals the Importance of Post-Transcriptional Regulation in Human Pluripotency
Investigators identified the protein interaction network of L1TD1 in human embryonic stem cells and provide insights into the interactome network constructed in human pluripotent cells. [Stem Cell Reports] Full Article | Graphical Abstract

Generation of Scaffoldless Hyaline Cartilaginous Tissue from Human iPSCs
The authors developed an approach to generate scaffoldless hyaline cartilage from human induced pluripotent stem cells (hiPSCs). Rowan Researcher Targets Stem Cell-Based Therapy for Rare Childhood Disease
Paola Leone, PhD, the director of the Cell and Gene Therapy Center and a professor of Cell Biology at the Rowan University School of Osteopathic Medicine, has been awarded a three-year, $477,000 grant from the National Institute of Neurological Disorders and Stroke to develop a stem cell-based therapy for Canavan disease.
