Inheritance of a Nuclear PIWI from Pluripotent Stem Cells by Somatic Descendants Ensures Differentiation by Silencing Transposons in Planarian
Using Dugesia japonica, the authors showed that a nuclear PIWI, DjPiwiB, was bound to PIWI-interacting RNAs (generally key mediators of PIWI-dependent transposon silencing), and was detected in not only neoblasts but also their descendant somatic cells, which do not express piwi. An Epigenetic Switch Regulates De Novo DNA Methylation at a Subset of Pluripotency Gene Enhancers during Embryonic Stem Cell Differentiation
Scientists investigated the effect of the Lsd1-Mi2/NuRD complex on the dynamic regulatory switch that induces the local interaction of histone tails with the Dnmt3 ATRX-DNMT3-DNMT3L domain, thus promoting DNA methylation at the enhancers of a subset of pluripotency genes. Tet Enzymes Regulate Telomere Maintenance and Chromosomal Stability of Mouse ESCs

Genome Therapy of Myotonic Dystrophy Type 1 iPS Cells for Development of Autologous Stem Cell Therapy
Investigators developed genome therapy in human myotonic dystrophy type 1 induced pluripotent stem (iPS) cells to eliminate mutant transcripts and reverse the phenotypes for developing autologous stem cell therapy. [Mol Ther] Abstract

Rapid Induction of Cerebral Organoids From Human Induced Pluripotent Stem Cells Using a Chemically Defined Hydrogel and Defined Cell Culture Medium
Researchers described a robust and simple method for generating cerebral organoids from human pluripotent stem cells by using a chemically defined hydrogel material and chemically defined culture medium. [Stem Cells Transl Med] Abstract

Generation and Gene Expression Profiling of 48 Transcription-Factor-Inducible Mouse Embryonic Stem Cell Lines
Previously, the authors generated and analyzed 137 transcription factor (TF)-inducible mouse embryonic stem cells (ESCs) lines. As an extension of this “NIA Mouse ESC Bank,” they generated and characterized 48 additional mouse ESC lines, in which single TFs in each line could be induced in a doxycycline-controllable manner. Generation and Characterization of Integration-Free Induced Pluripotent Stem Cells from Patients with Autoimmune Disease

Multiple Sclerosis Patient-Specific Primary Neurons Differentiated from Urinary Renal Epithelial Cells via Induced Pluripotent Stem Cells
Renal proximal tubule epithelial cells were isolated from multiple sclerosis patients’ urine and transfected with pluripotency-inducing episomal factors. Subsequent induced pluripotent stem cells were formed into embryoid bodies selective for ectodermal lineage, resulting in neural tube-like rosettes and eventually neural progenitor cells.
