    TOP STORY

MPP8 Is Essential for Sustaining Self-Renewal of Ground-State Pluripotent Stem Cells

Scientists identified the epigenetic protein MPP8 as being essential for ground-state pluripotency. Its depletion led to cell cycle arrest and spontaneous differentiation.

Study of Inherited Thrombocytopenia Resulting from Mutations in ETV6 or RUNX1 Using a Human Pluripotent Stem Cell Model

Utilizing CRISPR-Cas9, researchers compared the in vitro phenotype of hematopoietic progenitor cells and megakaryocytes derived from iPSC lines harboring mutations in either ETV6 or RUNX1.

Human iPSC-Based Neurodevelopmental Models of Globoid Cell Leukodystrophy Uncover Patient- And Cell Type-Specific Disease Phenotypes

Scientists investigated the impact of β-galactocerebrosidase (GALC) deficiency and lentiviral vector-mediated GALC rescue/overexpression in iPSC-derived neural progenitors and neuronal/glial progeny obtained from two globoid cell leukodystrophy patients.

Wnt–and Glutamate-Receptors Orchestrate Stem Cell Dynamics and Asymmetric Cell Division

Researchers discovered crosstalk between AMPA/Kainate-type ionotropic glutamate receptors and the Wnt-pathway during the initial Wnt3a-interaction at the cytonemes of mouse ESCs. They demonstrated that this crosstalk persisted throughout the Wnt3a-response in ESCs.

    Abstract

The Pluripotent Stem Cell-Specific Transcript ESRG Is Dispensable for Human Pluripotency

The global transcriptome data of various tissues and cell lines and quantitative expression analysis of PSCs showed that ESRG expression was much higher than other human endogenous retrovirus type-Hs and tightly silenced after differentiation.

Development, Characterization, and Hematopoietic Differentiation of Griscelli Syndrome Type 2 Induced Pluripotent Stem Cells

Researchers generated iPSCs from three different GS-2 patients using lentiviral vectors. The iPSCs were characterized using flow cytometry and RT-PCR and tested for the expression of pluripotency markers. In vivo differentiation to cells from all three germlines was tested using a teratoma assay.

Two Enhancers Regulate HoxB Genes Expression during Retinoic Acid-Induced Early Embryonic Stem Cells Differentiation through Long-Range Chromatin Interactions

CRISPR/Cas9-mediated individual or compound deletion of the two enhancers significantly inhibited HoxB gene expression, and transcriptome analysis revealed that retinoic acid-induced early ESCs differentiation was blocked in the enhancer KO cells.

Simplification of Culture Conditions and Feeder-Free Expansion of Bovine Embryonic Stem Cells

Simplified conditions will facilitate the use of bovine ESCs for gene editing applications and pluripotency and lineage commitment studies.
