    TOP STORY

Embryonic Origins of Adult Pluripotent Stem Cells

Researchers used Hofstenia miamia, a regenerative worm that possesses adult PSCs called neoblasts and produces manipulable embryos. Lineage tracing and functional experiments revealed that one pair of blastomeres gave rise to cells that resembled neoblasts in distribution, behavior, and gene expression.

An Optogenetic-Controlled Cell Reprogramming System for Driving Cell Fate and Light-Responsive Chimeric Mice

A light-inducible cell reprogramming system was developed based on a photoreceptor protein cryptochrome system and clustered regularly interspaced short palindromic repeats/nuclease-deficient CRISPR-associated protein 9 for iPSC reprogramming.

RNA Degradation Eliminates Developmental Transcripts during Murine Embryonic Stem Cell Differentiation via CAPRIN1-XRN2

To identify non-nuclear regulators of ESCs, scientists screened an endogenously labeled fluorescent fusion-protein library in mESCs. Cell-cycle-associated protein 1 (CAPRIN1) knockout had little effect in ESCs, but it significantly altered differentiation and gene expression programs.

KLF4 Inhibits Early Neural Differentiation of ESCs by Coordinating Specific 3D Chromatin Structure

Investigators observed enrichment of Krüppel-like factor 4 (KLF4) at some poised or closed enhancers at transcription start site-linked regions of genes associated with neural differentiation. Combination analysis of ChIP, HiChIP, and RNA-seq data indicated that KLF4 loss in ESCs induced changes in 3D chromatin structure.

The Epitope Basis of Embryonic Stem Cell-Induced Antitumor Immunity against Bladder Cancer

The authors tested prophylactic and therapeutic bladder cancer vaccines composed of allogeneic ESCs and CpG with or without GM-CSF.

Stress-Induced Reversible Cell-Cycle Arrest Requires PRC2/PRC1-Mediated Control of Mitophagy in Drosophila Germline Stem Cells and Human iPSCs

Scientists showed that mitophagy was a prerequisite for reversible quiescence in both irradiated Drosophila germline stem cells and hiPSCs.

Volumetric Imaging of Fast Cellular Dynamics with Deep Learning Enhanced Bioluminescence Microscopy

To increase the versatility of bioluminescence microscopy, investigators presented an improved low-light microscope in combination with deep learning methods to image extremely photon-starved samples enabling subsecond exposures for timelapse and volumetric imaging.

Efficient Simultaneous Double DNA Knock-In in Murine Embryonic Stem Cells by CRISPR/Cas9 Ribonucleoprotein-Mediated Circular Plasmid Targeting for Generating Gene-Manipulated Mice

Researchers first compared the knock-in (KI) efficiency of mESCs with CRISPR/Cas9 expression vector and ribonucleoprotein (RNP), and confirmed that KI efficiency was significantly increased by using RNP.

Requirement for STAT3 and Its Target, TFCP2L1, in Self-Renewal of Naïve Pluripotent Stem Cells In Vivo and In Vitro

The authors explored the requirement for the downstream signaling transducer and activator of transcription, STAT3, and its target, TFCP2L1, in the maintenance of naïve pluripotency.
