    TOP STORY

mRNA Translational Specialization by RBPMS Presets the Competence for Cardiac Commitment in hESCs

Researchers revealed that the competence for the future cardiac fate of hESCs was preset in pluripotency by a specialized mRNA translation circuit controlled by RNA binding protein with multiple splicing.

Preferential Hematopoietic Differentiation in Induced Pluripotent Stem Cells Derived from Human Umbilical Cord Arterial Endothelial Cells

hiPSCs were generated from somatic cells by transduction of 4 human transcription factors using Sendai virus vectors. Genome-wide DNA methylation analysis and transcriptional analysis were performed to evaluate the pluripotent capacity and somatic memory state of hiPSCs.

Generation of Functional Thymic Organoids From Human Pluripotent Stem Cells

Researchers showed the generation of thymic epithelial cells in vitro in isogenic stem cell-derived thymic organoids, consisting of thymic epithelial progenitors, hematopoietic progenitor cells, and mesenchymal cells, differentiated from the same hPSC line.

Generation of Hypothalamic Neural Stem Cell-Like Cells In Vitro from Human Pluripotent Stem Cells

Scientists generated hypothalamic neural stem cell-like cells from hESCs. They differentiated retina and anterior neural fold homeobox (RAX)::VENUS knockin hESCs into hypothalamic organoids and sorted RAX+ cells from mature organoids.

Rif1 Regulates Self-Renewal and Impedes Mesendodermal Differentiation of Mouse Embryonic Stem Cells

Investigators generated a Rif1 conditional knockout mESC line based on Cre-loxP system. Western blot, flow cytometry, qRT-PCR, RNA-Seq, ChIP-Seq, ChIP-qPCR, immunofluorescence, and immunoprecipitation were employed for phenotype and molecular mechanism assessment.

Commercial Dishes with Gelatin-Free Microstructured Inserts for Elongated Stem Cell Self-Renewal and Pluripotency

The authors reported scalable fabrication of 2i-functionalized micro-pyramid-array inserts for use in commercial multiwell plates, as the alternative cultivation platform for maintaining long-term self-renewal and pluripotency of multiple mESCs and miPSCs.

The Enhanced Generation of Motor Neurons from mESCs by MgAl Layered Double Hydroxide Nanoparticles

Researchers studied the effects of layered double hydroxide (LDH) without loaded factors on mESC neurogenesis. The enhanced differentiation of mESCs into motor neurons by LDH was systematically validated by immunofluorescent staining, qRT-PCR analysis, and western blot analysis.

Predicting Reprogramming-Related Gene Expression from Cell Morphology in Human Induced Pluripotent Stem Cells

Scientists investigated the relationship between gene expression levels and morphology by analyzing live-cell phase-contrast images and mRNA profiles collected during the purification process.

    Abstract

Genetic Dissection of the Pluripotent Proteome through Multi-Omics Data Integration

Investigators performed a comprehensive proteogenomics analysis of 190 genetically diverse mESC lines. The quantitative proteome was highly variable across lines, and they identified pluripotency-associated pathways that were differentially activated in the proteomics data.
