Crosstalk between RNA m6A and DNA Methylation Regulates Transposable Element Chromatin Activation and Cell Fate in Human Pluripotent Stem Cells

Researchers developed a targeted proteomic proximity labeling approach to capture transposable element (TE)-associated proteins in hESCs. They found that the RNA N6-methyladenosine (m6A) reader, YTHDC2, occupied the genomic loci of the primate-specific TE, LTR7/HERV-H.

Functional Annotation of Variants of the BRCA2 Gene via Locally Haploid Human Pluripotent Stem Cells

By using Cas9 to edit the functional BRCA2 allele in the locally haploid hPSCs and in fibroblasts differentiated from them, scientists characterized essential regions in the gene to identify permissive and loss-of-function mutations.

A Genome-Wide Screen Reveals New Regulators of the 2-Cell-Like Cell State

To better understand totipotency, investigators carried out a genome-wide CRISPR knockout screen in mouse ESCs, searching for mutants that reactivate the expression of Dazl, a gene expressed in 2-cell-like cells.

WD Repeat Domain 82 (Wdr82) Facilitates Mouse iPSCs Generation by Interfering Mitochondrial Oxidative Phosphorylation and Glycolysis

On the basis of Yamanaka factors OSKM induction method, MEF cells were induced and reprogrammed into iPSCs under conditions of the oocyte-derived factor Wdr82 overexpression and/or knockdown, so as to assess the reprogramming efficiency.

Multi-Level Fgf4- and Apoptosis-Dependent Regulatory Mechanism Ensures the Plasticity of ESC-Chimeric Mouse Embryo

Using mouse chimeric embryos, constructed by injection of ESCs into the eight-cell embryo, as a tool, scientists aimed to explore the mechanisms underlying the regulatory nature of the preimplantation mouse embryo.

Zfp296 Knockout Enhances Chromatin Accessibility and Induces a Unique State of Pluripotency in Embryonic Stem Cells

Transcriptome analysis revealed that Zfp296 deficiency in Zfp296-knockout ESCs up- and down-regulated a distinct group of genes, among which Dppa3, Otx2, and Pou3f1 were markedly downregulated.

Network Characterization linc1393 in the Maintenance of Pluripotency Provides the Principles for lncRNA Targets Prediction

Researchers demonstrated that a nuclear lncRNA linc1393 maintained mouse ESC pluripotency by recruiting SET1A near its binding sites, to establish H3K4me3 status and activate the expression of specific pluripotency-related genes.

Human IP3 Receptor Triple Knockout Stem Cells Remain Pluripotent Despite Altered Mitochondrial Metabolism

Given the importance of inositol 1,4,5-trisphosphate receptors (IP3Rs) in human diseases, the authors investigated their role in hiPSC by developing single IP3R and triple IP3R knockouts.
