Durable Alveolar Engraftment of PSC-Derived Lung Epithelial Cells into Immunocompetent Mice

Researchers differentiated murine PSCs into self-renewing lung epithelial progenitors able to engraft into the injured distal lung epithelium of immunocompetent, syngeneic mouse recipients.

The Histone Chaperone Function of Daxx Is Dispensable for Embryonic Development

To advance our understanding of histone chaperone activity in vivo, scientists developed two Daxx mutant alleles in the mouse germline, which abolish the interactions between Daxx and Atrx, and Daxx and H3.3

Haspin Balances the Ratio of Asymmetric Cell Division Through Wnt5a and Regulates Cell Fate Decisions in Mouse Embryonic Stem Cells

The authors reported that Haspin knockout mouse (m)ESCs had increased ratio of asymmetric cell division, which cumulatively regulated cell fate decisions. Furthermore, Wnt5a was significantly downregulated due to decreased Pax2 in Haspin-KO mESCs.

Effects of Low Doses of Methylmercury (MeHg) Exposure on Definitive Endoderm Cell Differentiation in Human Embryonic Stem Cells

Investigators determined the effects of low concentrations of MeHg exposure during the differentiation of definitive endoderm (DE) cells from human ESCs. hESCs were exposed to MeHg that covered the range of Hg concentrations typically found in human maternal blood during DE cell induction.

Production and Limbal Lineage Commitment of Aniridia Patient-Derived Induced Pluripotent Stem Cells

The authors produced hiPSC from three aniridia-associated keratopathy patients and examined whether PAX6 haploinsufficiency affected limbal stem cell lineage commitment.

Improved Cryopreservation of Human Induced Pluripotent Stem Cell (iPSC) and iPSC Derived Neurons Using Ice-Recrystallization Inhibitors

Scientistss investigated the role of ice recrystallization inhibitors (IRIs) as novel cryoprotectants for iPSCs and terminally differentiated iPSC-Ns. They found that one class of IRIs, N-aryl-D-aldonamides, increased iPSC post-thaw viability and recovery with no adverse effect on iPSC pluripotency.

p62 and NBR1 Functions Are Dispensable for Aggrephagy in Mouse ESCs and ESC-Derived Neurons

Data revealed that both ESCs and ESC-derived neurons did not show strong defects in the clearance of protein aggregates upon knockout of p62 or NBR1 and upon mutation of the p62 ubiquitin-associated domain and the LC3-interacting region motif.

The Induction of Parathyroid Cell Differentiation from Human Induced Pluripotent Stem Cells Promoted via TGF-α/EGFR Signaling

Investigators developed a new method of parathyroid cell differentiation from hiPS cells. The parathyroid cell differentiation occurred in accordance with embryologic development.

    Abstract

Wnt and BMP Signaling Direct Anterior/Posterior Differentiation in Aggregates of Mouse Embryonic Stem Cells

Researchers showed that Wnt signaling had a general posteriorising effect on stem cell aggregates and directed differentiation towards the mesoderm, as confirmed through the upregulation of posterior and mesodermal markers.

Control of Stem Cell Renewal and Fate by YAP and TAZ

YAP and TAZ translate complex biological cues acting on stem cells — from mechanical forces to cellular metabolism — into genome-wide effects to mediate stem cell functions.

Single-Molecule Tracking of Nanog and Oct4 in Living Mouse Embryonic Stem Cells Uncovers a Feedback Mechanism of Pluripotency Maintenance

To understand their function in the pluripotency maintenance, researchers visualised and quantified the dynamics of single molecules of Nanog and Oct4 in a mouse ESCs during pluripotency loss.
