    TOP STORY

Modeling Post-Implantation Human Development to Yolk Sac Blood Emergence

Investigators presented a genetically inducible stem cell-derived embryoid model of early post-implantation human embryogenesis that captured the reciprocal co-development of embryonic tissue and extra-embryonic endoderm and mesoderm niche with early hematopoiesis.

XIST Directly Regulates X-Linked and Autosomal Genes in Naive Human Pluripotent Cells

The authors found that XIST spread across the X chromosome and induced dampening of X-linked gene expression in naive hPSCs. Surprisingly, XIST also targets specific autosomal regions, where it induces repressive chromatin changes and gene expression dampening.

FOXO1-Mediated Lipid Metabolism Maintains Mammalian Embryos in Dormancy

Scientists showed that mouse embryos in diapause were sustained by using lipids as primary energy source. In vitro, supplementation of embryos with the metabolite L-carnitine balances lipid consumption, put the embryos in deeper dormancy and boosted embryo longevity.

Amino Acid Intake Strategies Define Pluripotent Cell States

Researchers showed that mouse ESCs captured the unique metabolic state of preimplantation embryos and proliferated in the absence of several essential amino acids.

    Abstract

Rapid Human Genomic DNA Cloning into Mouse Artificial Chromosome via Direct Chromosome Transfer From Human iPSC and CRISPR/Cas9-Mediated Translocation

Scientists achieved a direct transfer of human chromosome 6 or 21 as a model from hiPSCs as alternative human chromosome donors into CHO cells containing mouse artificial chromosome.

DNA Methylation Restricts Coordinated Germline and Neural Fates in Embryonic Stem Cell Differentiation

Using culture modeling of cellular transitions, researchers found that DNA methylation-free mouse ESCs with triple DNA methyltransferase knockout progressed through the continuum of pluripotency states but demonstrated skewed differentiation abilities toward neural versus other somatic lineages.

Mitotic Bookmarking Redundancy by Nuclear Receptors in Pluripotent Cells

The authors used targeted proteomics of the mitotic bookmarking transcription factor ESRRB, an orphan nuclear receptor, to discover a large redundancy in mitotic binding among members of the protein super-family of nuclear receptors.

Scalable Production of Homogeneous Cardiac Organoids Derived from Human Pluripotent Stem Cells

Investigators described a large-scale method for producing homogeneous hiPSC-derived cardiac spheroids and cardiac organoids without scaffolds using a porous 3D microwell substratum with a suction system.

Metabolic Memory of Δ9-Tetrahydrocannabinol Exposure in Pluripotent Stem Cells and Primordial Germ Cells-Like Cells

Researchers leveraged a stepwise in vitro differentiation system that captured the early embryonic developmental cascade to investigate the impact of exposure to the most abundant phytocannabinoid, Δ9-tetrahydrocannabinol.

    Abstract

In Vitro Production of Naked Mole-Rats’ Blastocysts From Non-Breeding Females Using In Vitro Maturation and Intracytoplasmic Sperm Injection

Immature oocytes, opportunistically retrieved from sexually inactive females, underwent first in vitro maturation and then in vitro fertilization via piezo-intracytoplasmic sperm injection.

The Stem Cell Zoo for Comparative Studies of Developmental Tempo

Investigators discuss how PSC-based models of development can be used to investigate cell- and tissue-autonomous temporal processes.
