Two-Way Regulation of MgFe-LDH on ESC Cell Fate via Temporal Selective Activation of Different Cell Membrane Receptors

Investigators revealed that MgFe-layered double hydroxide (LDH) had bidirectional regulatory effects on the differentiation of mESCs into neural progenitor cells, essentially suppressing cell differentiation on the first day and promoting cell differentiation on the third day.

Annelid Adult Cell Type Diversity and Their Pluripotent Cellular Origins

Researchers recovered 75,218 single cell transcriptomes of the highly regenerative and asexually-reproducing annelid Pristina leidyi.

Cell-Type-Specific mRNA Transcription and Degradation Kinetics in Zebrafish Embryogenesis from Metabolically Labeled Single-Cell RNA-Seq

Scientists combined single-cell RNA-Seq and metabolic labeling to capture temporal cellular transcriptomes of zebrafish embryos where newly-transcribed and pre-existing mRNA could be distinguished.

Smaug Regulates Germ Plasm Assembly and Primordial Germ Cell Number in Drosophila Embryos

Investigators identified mechanisms that subsequently regulated germ plasm assembly in the early embryo. They showed that the Smaug RNA binding protein was transported into the germ plasm of the early embryo where it accumulated in the germ granules.

TEAD2 Initiates Ground-State Pluripotency by Mediating Chromatin Looping

Researchers systematically screened factors essential for ESC pluripotency, identifying transcriptional enhanced associate domain (TEAD) 2 as an ancillary factor maintaining ground-state pluripotency in 2i/leukemia inhibitory factor (LIF) ESCs and facilitating the transition from serum/LIF to 2i/LIF ESCs.

Basal Delamination during Mouse Gastrulation Primes Pluripotent Cells for Differentiation

The authors developed and characterized a 3D culture of self-renewing mouse embryonic cells that captured the main transcriptional and architectural features of the early gastrulating mouse epiblast.

Low Initial Cell Density Promotes the Differentiation and Maturation of Human Pluripotent Stem Cells into Erythrocytes

In the selection of low- and high-density inoculation conditions, scientists found that low-density culture performed better in the final red blood cell product with more cell output and increased average cellular hemoglobin content.

Impaired Osteogenesis in Human Induced Pluripotent Stem Cells with Acetaldehyde Dehydrogenase 2 Mutations

Researchers investigated the effects of acetaldehyde dehydrogenase 2 (ALDH2) mutations on bone remodeling. They examined the effects of ALDH2 polymorphisms on in vitro osteogenesis using hiPSCs.

Human Embryonic Genetic Mosaicism and Its Effects on Development and Disease

Recent high-throughput sequencing studies have revealed that mosaicism is common, clone sizes often increase with age and specific variants can affect tissue function and disease development.

The Many Faces of H3.3 in Regulating Chromatin in Embryonic Stem Cells and Beyond

The authors provide an update with a focus on H3.3 functions in early mammalian development, considering the context of ESC maintenance and differentiation, to finally conclude with emerging roles in cancer development and cell fate transition and maintenance.
