Genetic and Chemical Targeting of the ATPase Complex TIP48 and 49 Impairs Acute Myeloid Leukemia

Researchers showed that loss of Tip49 triggered p53-dependent apoptosis and inhibited leukemia development in vivo. To examine the impact of chemical inhibition of this complex on leukemia, they developed the novel compound DS-4950, which interfered with the ATPase activity of the Tip48/49.

Activation of GPR44 Decreases Severity of Myeloid Leukemia via Specific Targeting of Leukemia Initiating Stem Cells

Scientists showed that AML leukemia initiating stem cells were sensitive to endogenous and exogenous cyclopentenone prostaglandin-J, Δ12-PGJ2, and 15d-PGJ2, which were increased upon dietary selenium supplementation via the cyclooxygenase-hematopoietic PGD synthase pathway.

ARHGAP4 Promotes Leukemogenesis in Acute Myeloid Leukemia by Inhibiting DRAM1 Signaling

Investigators studied lymphoma/leukemia-related activity in the evolving landscape of cells’ commitment to the erythroid lineage and specifically during fetal to adult hemoglobin transversion, to qualify transcription factors affecting chromatin remodeling as potential repressors of lncRNAs and miRNAs.

Ciclopirox Ethanolamine Preserves the Immature State of Human HSCs by Mediating Intracellular Iron Content

Scientists found that the antifungal agent ciclopirox ethanolamine selectively supported immature CD34+CD90+ cells during culture and enhanced their long-term in vivo repopulation capacity.

Loss of Nudt15 Thiopurine Detoxification Increases Direct DNA Damage in Hematopoietic Stem Cells

Scientists investigated the effects of nucleoside diphosphate-linked moiety X-type motif 15 (NUDT15) R139C on hematopoietic stem cells upon thiopurine administration.

Transcriptional Repression of lncRNA and miRNA Subsets Mediated by Lrf During Erythropoiesis

Researchers showed that deubiquitinase USP9X stabilized ALKBH5 and promoted AML cell survival. Through the use of mass spectrometry as an unbiased approach, they identified USP9X and confirmed that it directly bound to ALKBH5.

Analysis of Self-Renewing and Differentiation-Related microRNAs and Transcription Factors in Multilineage Mouse Hematopoietic Stem/Progenitor Cells Induced by 1,4-Benzoquinone

The effect of a benzene metabolite, 1,4-benzoquinone exposure, in hematopoietic stem and progenitor cells focusing on the self-renewing and differentiation pathways was investigated.

Acute Megakaryoblastic Leukemia Shows High Frequency of Chromosome 1q Aberrations and Dismal Outcome

Scientists performed genetic analyses, and evaluated prognostic factors and the value of allogeneic hematopoietic stem cell transplantation in a homogenous adult acute megakaryoblastic leukemia patient cohort.

Influence of Conditioning Regimen Intensity on Outcomes Post-Allogeneic Hematopoietic Cell Transplantation for Acute Myeloid Leukemia in Complete Morphological Remission

Investigators retrospectively analyzed 451 patients who underwent allogeneic hematopoietic cell transplantation for AML in complete remission with either reduced intensity conditioning or myeloablative with the use of dual T cell depletion as graft-versus-host disease prophylaxis.
