    TOP STORY
IL8-CXCR2 Pathway Inhibition as a Therapeutic Strategy against MDS and AML Stem Cells
Investigators conducted parallel transcriptional analysis of highly fractionated stem and progenitor populations in MDS, AML and control samples and found interleukin 8 (IL8) to be consistently overexpressed in patient samples. [Blood] Abstract

PUBLICATIONS (Ranked by impact factor of the journal)
Loss of Dnmt3a and Endogenous KrasG12D/+ Cooperate to Regulate Hematopoietic Stem and Progenitor Cell Functions in Leukemogenesis
Researchers found that Dnmt3a-/- induces multiple hematopoietic phenotypes after a prolonged latency, including T cell expansion in peripheral blood, stress erythropoiesis in spleen, and myeloid malignancies in liver. [Leukemia] Abstract

Induction of Hematopoietic and Endothelial Cell Program Orchestrated by ETS Transcription Factor ER71/ETV2
Scientists mapped ETV2 global binding sites and carry out in vitro differentiation of embryonic stem cells, and germ line and conditional knockout mouse studies to uncover mechanisms involved in the hemangiogenic fate commitment from mesoderm. [EMBO Rep] Abstract | Graphical Abstract

EphB2 and EphB3 Play an Important Role in the Lymphoid Seeding of Murine Adult Thymus
Adult thymuses lacking either ephrin type B receptor 2 (EphB2) or EphB3, or expressing a truncated form of EphB2, the forward signal-deficient EphB2LacZ, have low numbers of early thymic progenitors and are colonized in vivo by reduced numbers of injected bone marrow lineage-negative cells. Hematopoietic progenitors from these EphB mutants showed decreased capacities to colonize wild type (WT) thymuses compared with WT precursors, with EphB2-/- cells exhibiting the greatest reduction. [J Leukoc Biol] Abstract

Detecting Primitive Hematopoietic Stem Cells in Total Nucleated and Mononuclear Cell Fractions from Umbilical Cord Blood Segments and Units
63 umbilical cord blood (UCB) segments and 10 UCB units plus segments were analyzed for the response of both primitive lympho-hematopoietic and primitive hematopoietic stem cells in both the total nucleated cell and mononuclear cell fractions. [J Transl Med] Full Article

Wnt3a Protein Reduces Growth Factor-Driven Expansion of Human Hematopoietic Stem and Progenitor Cells in Serum-Free Cultures
Scientists demonstrated that exogenous Wnt3a protein suppresses rather than promotes the expansion of umbilical cord blood-derived CD34+ cells in serum free expansion cultures. [PLoS One] Full Article

Mitochondria Defects Are Involved in Lead-Acetate-Induced Adult Hematopoietic Stem Cell Decline
Investigators performed oral intragastric administration of various dose lead acetate to adult Sprague Dawley rats to define the role of lead exposure in hematopoietic stem cells function, and to clarify its underlying mechanism. [Toxicol Lett] Abstract

    CLINICAL RESEARCH

Allogeneic Stem Cell Transplantation May Improve the Outcome of Adult inv(16) Acute Myeloid Leukemia Patients in First Complete Remission with Poor Molecular Responses to Chemotherapy
86 adult inv(16) acute myeloid leukemia patients in the first complete remission were serially monitored for CBFB-MYH11 transcript levels during the early courses of chemotherapy. 57 and 29 of them received chemotherapy/autologous SCT and allo-SCT after the second consolidation, respectively. [Leuk Lymphoma] Abstract

Thiotepa-Based versus Total Body Irradiation-Based Myeloablative Conditioning Prior to Allogeneic Stem Cell Transplantation for Acute Myeloid Leukemia in First Complete Remission: A Retrospective Analysis from the Acute Leukemia Working Party of the European Group for Blood and Marrow Transplantation
Investigators evaluated the outcome of adults with acute myeloid leukemia in first complete remission who received myeloablative conditioning either with a thiotepa-based or a cyclophosphamide/total body irradiation-based regimen for allogeneic hematopoietic stem cell transplantation from an HLA-matched sibling or an unrelated donor. [Eur J Haematol] Abstract

    REVIEWS
Advances and Challenges in Immunotherapy for Solid Organ and Hematopoietic Stem Cell Transplantation
The authors outline the current immunological limitations for hematopoietic stem cell and solid organ transplantation and discuss new immune-modulating therapies in preclinical development and in clinical trials that may allow these obstacles to be overcome. [Sci Transl Med] Abstract

Cellular Innate Immunity and Restriction of Viral Infection – Implications for Lentiviral Gene Therapy in Human Hematopoietic Cells
The authors summarize the current knowledge regarding innate immune recognition of lentiviruses in primary human hematopoietic cells as well as discuss its relevance for lentiviral vector-based ex-vivo gene therapy approaches. [Hum Gene Ther] Abstract
