    TOP STORY
Adenosine Signaling Promotes Hematopoietic Stem and Progenitor Cell Emergence
Researchers showed that adenosine signaling regulates hematopoietic stem and progenitor cell (HSPC) development in zebrafish embryos. The adenosine receptor A2b is expressed in the vascular endothelium before HSPC emergence. [J Exp Med] Abstract
Development of Simplified and Defined Cell Culture for hPSCs - Watch Now!

PUBLICATIONS (Ranked by impact factor of the journal)
Loss of TET2 in Hematopoietic Cells Leads to DNA Hypermethylation of Active Enhancers and Induction of Leukemogenesis
Scientists present a novel Tet2-dependent leukemia mouse model that closely recapitulates gene expression profiles and hallmarks of human AML1-ETO-induced acute myeloid leukemia. [Genes Dev] Abstract

Wip1 Deficiency Impairs Hematopoietic Stem Cell Function via p53 and mTORC1 Pathways
Researchers showed that wild-type p53-induced phosphatase 1 (Wip1) is highly expressed in hematopoietic stem cells (HSCs) but decreases with age. Wip1-deficient mice exhibited multifaceted HSC aging phenotypes, including the increased pool size and impaired repopulating activity. [Nat Commun] Abstract

Targeting of Folate Receptor-Beta on Acute Myeloid Leukemia Blasts with Chimeric Antigen Receptor Expressing T Cells
As many cell-surface markers are shared between acute myeloid leukemia blasts and healthy hematopoietic stem and progenitor cells (HSCs), investigators evaluated folate receptor β expression and recognition of HSCs by chimeric antigen receptor T cells. [Blood] Abstract

Macrophage-Lineage Cells Negatively Regulate the Hematopoietic Stem Cell Pool in Response to IFNγ at Steady State and during Infection
Scientists demonstrated that depletion of bone marrow resident macrophages increases hematopoietic stem cell (HSC) proliferation as well as the pool of quiescent HSCs. [Stem Cells] Abstract

hCD2-iCre and Vav-iCre Mediated Gene Recombination Patterns in Murine Hematopoietic Cells
To better characterize which hematopoietic cells contain hCD2-iCre activity, investigators compared EYFP fluorescence in hCD2-iCre+/- R26-stop-EYFP+/- and Vav-iCre+/- R26-stop-EYFP+/- mice. [PLoS One] Full Article

    CLINICAL RESEARCH

Randomized Study of Reduced-Intensity Chemotherapy Combined with Imatinib in Adults with Ph-Positive Acute Lymphoblastic Leukemia
Scientists randomly compared high doses of the tyrosine kinase inhibitor imatinib combined with reduced-intensity chemotherapy to standard imatinib/HyperCVAD treatment in 268 adults with Ph-positive acute lymphoblastic leukemia. [Blood] Abstract

Comparison of Outcomes after Unrelated Cord Blood and Unmanipulated Haploidentical Stem Cell Transplantation in Adults with Acute Leukemia
Investigators compared outcomes after unrelated cord blood transplantation and haploidentical stem cell transplantation in adults with de novo acute myeloid leukemia and acute lymphoblastic leukemia. [Leukemia] Abstract

Does Defibrotide Induce a Delay to Polymorphonuclear Neutrophil Engraftment after Hematopoietic Stem Cell Transplantation? Observation in a Pediatric Population
Researchers investigated whether defibrotide (DFT) prophylaxis affects neutrophil engraftment in patients undergoing hematopoietic stem cell transplantation (HSCT). A cohort of 44 consecutive pediatric patients who underwent HSCT was retrospectively analyzed to see the role of DFT on engraftment. [Adv Ther] Abstract
