PUBLICATIONS(Ranked by impact factor of the journal)Researchers describe a Treg subpopulation that controls HSC quiescence and engraftment. These Tregs highly expressed an HSC marker, CD150, and localized within the HSC niche in the bone marrow. [Cell Stem Cell] Abstract | Graphical Abstract Investigators demonstrated a crucial role for STAT3 antiinflammatory activity in preservation of hematopoietic stem and progenitor cells (HSPCs) and lineage-balanced hematopoiesis. Conditional Stat3 removal from the hematopoietic system led to depletion of the bone marrow lineage− Sca-1+ c-Kit+ CD150+ CD48− HSPC subset, myeloid-skewed hematopoiesis, and accrual of DNA damage in HSPCs. [Proc Natl Acad Sci USA] Abstract Lineage-Biased Hematopoietic Stem Cells Are Regulated by Distinct Niches Researchers showed that platelet and myeloid-biased HSCs, marked by von Willebrand factor (vWF) expression, are highly enriched in megakaryocytes (MK) niches. Depletion of MK selectively expanded vWF+ HSCs, whereas the depletion of NG2+ arteriolar niche cells selectively depletes vWF− lymphoid-biased HSCs. [Dev Cell] Full Article | Graphical Abstract ERα Promotes Murine Hematopoietic Regeneration through the Ire1α-Mediated Unfolded Protein Response Scientists showed that the female sex hormone estrogen regulates the unfolded protein response in HSCs. Estrogen treatment increases the capacity of HSCs to regenerate the hematopoietic system upon transplantation and accelerates regeneration after irradiation. [Elife] Full Article Using knockout mice, investigators demonstrated that bromodomain PHD finger transcription factor (BPTF), a component of the nucleosome remodeling factor chromatin-remodeling complex, is essential for maintaining the population size of hematopoietic stem/progenitor cells, including long-term HSCs. [Stem Cell Reports] Full Article Murine Hematopoietic Stem Cell Reconstitution Potential Is Maintained by Osteopontin during Aging Investigators studied the impact of osteopontin (OPN), on HSC function during aging using an OPN-knockout mouse model. They showed that during aging OPN deficiency is associated with an increase in lymphocytes and a decline in erythrocytes in peripheral blood. [Sci Rep] Full Article Researchers compared the sensitivity of HSCs with the genotoxicity testing cell line TK6 for chromosomal mutations. HSCs were less sensitive than TK6 cells for the genotoxic effects of the model genotoxins and chemotherapeutic agents doxorubicin, vinblastine, methyl methanesulfonate and equally sensitive for mitomycin C. [Sci Rep] Full Article The authors identified miR-193a and miR-600 targeting and degrading Wilm’s tumor-1 (WT1). Ectopic expressions of miR-193a and miR-600 synergistically inhibited cell proliferation, induced apoptosis, and decreased colony formation in leukemia cells. [Exp Hematol] Abstract | Graphical Abstract CLINICAL RESEARCHResearchers profiled miR-193b-5p/3p expression in cytogenetically and clinically characterized de novo pediatric acute myeloid leukemia (AML) via quantitative real-time polymerase chain reaction and validated their findings in an independent cohort of 187 adult patients. They investigated the tumor suppressive function of miR-193b in human AML blasts, patient-derived xenografts, and miR-193b knockout mice in vitro and in vivo. [J Clin Oncol] Full Article | |

POLICY NEWSBiden Blasts Science Denialists, Calls to Dramatically Speed Up Fight against Cancer Fresh off a plane from the NATO meeting in Munich, where he’d conferred with world leaders on global defense, former U.S. Vice President Joe Biden issued a forceful call here last night to build on the “Cancer Moonshot” he started while in the White House and transform the way the world fights cancer. [ScienceInsider] Editorial Indonesian Scientists Hamstrung by Year-Long Funding Delay Indonesian scientists expecting to receive hundreds of thousands of dollars for research are stuck in limbo because the country’s first dedicated science-funding agency is struggling to raise enough money. [Nature News] Editorial Italian Election Leaves Science out in the Cold As campaigning ahead of Italy’s national election enters its final weeks, researchers in the country fear that budget cuts and declining interest in science will only continue.
