TOP STORYComplex Mammalian-Like Hematopoietic System Found in a Colonial Chordate Scientists describe the hematopoietic system of Botryllus schlosseri, a colonial tunicate that has a vasculature and circulating blood cells, and interesting stem-cell biology and immunity characteristics. Using flow cytometry, whole-transcriptome sequencing of defined cell populations and diverse functional assays, they identified HSCs, progenitors, immune effector cells and an HSC niche, and demonstrated that self-recognition inhibits allospecific cytotoxic reactions. Overexpression of Ars2 promoted cell proliferation and colony formation in AML cells, whereas depletion of Ars2 inhibited cell proliferation and colony formation. [Leukemia] Full Article Enhancing IGFBP7 by overexpression or addition of recombinant human IGFBP7 (rhIGFBP7) resulted in differentiation, inhibition of cell survival, and increased chemotherapy sensitivity of primary acute myeloid leukemia cells. Adding rhIGFBP7 reduced leukemic stem and/or progenitor survival and reversed a stem-like gene signature, but it had no influence on normal hematopoietic stem cell survival.
