Long-term stable cell lines were obtained from a hepatocellular carcinoma (HCC)-bearing rat exposed to the Resistant-Hepatocyte protocol and from a rat subjected to the same model in the absence of carcinogenic treatment, thus not developing HCC. [Oncotarget] Full Article Since hepatocellular carcinoma mostly develops on a cirrhotic background, the authors sought to determine whether cancer stem cell-like epithelial cell adhesion molecule (EpCAM)+ cells exist in patients with advanced cirrhosis. Both flow cytometry and immunohistochemistry showed that frequency of EpCAM+ cells in advanced cirrhosis was increased as compared to control. [Stem Cells Transl Med] Abstract In three-dimensional extracellular matrix culture, cysteine cathepsin (Cts)H, among other cysteine cathepsins, was up-regulated and localized as puncta in the nuclear and cytoplasmic compartments in a complex with histone deacetylase (HDAC)4 for its degradation. Conversely, along with hepatic stellate cell trans-differentiation, CtsH and CtsL were progressively down-regulated, whereas HDAC4 was concurrently stabilized. [Am J Pathol] Abstract NS5ATP13 Promotes Liver Fibrogenesis via Activation of Hepatic Stellate Cells Researchers found that HCV NS5A-transactivated protein 13 (NS5ATP13) was upregulated in both fibrotic liver tissues and activated human hepatic stellate cells (HSCs) induced by TGF-β1. Moreover, NS5ATP13 enhanced extracellular matrix production and HSC activation, with or without TGF-β1 treatment, likely involving the TGF-β1/Smad3 signaling pathway.
