PUBLICATIONS(Ranked by impact factor of the journal)Scientists observed that the elevated expression of both ZFX and epithelial cell adhesion molecule (EpCAM) was associated with aggressive clinicopathological features and indicated poor prognosis in patients with hepatocellular carcinoma. ZFX was commonly enriched in liver EpCAM+ cancer stem cells. [Mol Oncol] Abstract | Full Article Human iPSCs were generated from patients with bile salt export pump (BSEP)-deficiency (BD-iPSC), and were differentiated into hepatocyte-like cells (HLCs). In the BD-iPSC derived HLCs, BSEP was not expressed on the cell surface and the biliary excretion capacity was significantly impaired. [Sci Rep] Full Article Prostate-derived E-twenty-six (ETS) factor (PDEF) expression was suppressed in highly metastatic hepatocellular carcinoma (HCC) cell lines, and shRNA-mediated down-regulation of PDEF in low-metastatic HCC cell lines significantly increased cellular proliferative and invasive capacity in vitro and in vivo. [Oncotarget] Full Article Scientists describe the isolation and characterization of two rat hepatocyte cell lines as tools to study liver carcinogenesis. Long-term stable cell lines were obtained from a hepatocellular carcinoma (HCC)-bearing rat exposed to the Resistant-Hepatocyte protocol and from a rat subjected to the same model in the absence of carcinogenic treatment, thus not developing HCC. [Oncotarget] Full Article Since hepatocellular carcinoma mostly develops on a cirrhotic background, the authors sought to determine whether cancer stem cell-like epithelial cell adhesion molecule (EpCAM)+ cells exist in patients with advanced cirrhosis. Both flow cytometry and immunohistochemistry showed that frequency of EpCAM+ cells in advanced cirrhosis was increased as compared to control. [Stem Cells Transl Med] Abstract In three-dimensional extracellular matrix culture, cysteine cathepsin (Cts)H, among other cysteine cathepsins, was up-regulated and localized as puncta in the nuclear and cytoplasmic compartments in a complex with histone deacetylase (HDAC)4 for its degradation. Conversely, along with hepatic stellate cell trans-differentiation, CtsH and CtsL were progressively down-regulated, whereas HDAC4 was concurrently stabilized. [Am J Pathol] Abstract NS5ATP13 Promotes Liver Fibrogenesis via Activation of Hepatic Stellate Cells Researchers found that HCV NS5A-transactivated protein 13 (NS5ATP13) was upregulated in both fibrotic liver tissues and activated human hepatic stellate cells (HSCs) induced by TGF-β1. Moreover, NS5ATP13 enhanced extracellular matrix production and HSC activation, with or without TGF-β1 treatment, likely involving the TGF-β1/Smad3 signaling pathway. [J Cell Biochem] Abstract Investigators compared three emerging cell systems, hepatocytes derived from induced pluripotent stem cells, HepaRG cells and 3D primary human hepatocyte spheroids at transcriptional and functional levels in a multi-center study to evaluate their potential as predictive models for drug-induced hepatotoxicity. [Drug Metab Dispos] Abstract Highly Tumorigenic Hepatocellular Carcinoma Cell Line with Cancer Stem Cell-Like Properties The authors derived a new cell line from Hepa1-6 cells through an in vivo passage in C57BL/6 mice. The resulting Dt81Hepa1-6 cell line showed enhanced tumorigenicity compared to Hepa1-6 with more frequent and more rapid tumor development in C57BL/6 mice. [PLoS One] Full Article | |
